The research herein builds on the work of Rojek (2003) and further investigates the bioremediation of NOM wastes. The aim of this research was to evaluate the effectiveness of selected white-rot fungi and yeast for the biodegradative removal of concentrated NOM from solution. The effectiveness of the white-rot fungus P. chrysosporium ATCC 34541 for the biological degradation of different batches of MIEXTM NOM concentrate and the effects of different characteristics of the NOM on the process and thus the resistance of the NOM to microbial removal were investigated.

The purpose of this research was to investigate the technological feasibility of biological filtration process for Mn control in Lorne WTP. This thesis consists of eight chapters. Project background and research objectives are described in Chapter 1. Chapter 2 presents an extensive literature review covering the nature of manganese, manganese analytical methods, manganese control methods, the nature of MOB and biological control of Mn in drinking water treatment. The materials and methods used throughout the experimental investigation for this study are described in Chapter 3.

Slow sand filtration has been an effective means of treating water for the control of microbiological contaminants. Schmutzdecke is the layer of the microbial community that is responsible for treating the water through the sand bed. As water passes through this biological layer, foreign particles are trapped and essentially eaten by bacteria forms on this layer. Development of Schmutzdecke layer depends on available microbes in raw water, food supply, oxygen supply, residence time, and wetting of sand bed.

Trehalose synthase (TreS) catalyzes the reversible inter-conversion of trehalose (glucosyl-a,a-1,1-glucose) and maltose (glucosyl-a1-4-glucose). TreS was purified from the cytosol ofMycobacterium smegmatisto give a single protein band on SDS gels with a molecular mass of68 kDa. However, active enzyme exhibited a molecular mass of 390 kDa by gel filtration suggesting that TreS is a hexa-mer of six identical subunits.

The effect of pH and temperature on structure, stability, activity and enantioselectivity of haloalkane dehalogenase DbjA from Bradyrhizobium japonicum USDA110 was investigated in this study. Conformational changes have been assessed by circular dichroism spectroscopy, functional changes by kinetic analysis, while quaternary structure was studied by gel filtration chromatography.

Potato tuber storage proteins were obtained from vacuoles isolated from field-grown starch potato tubers cv. Kuras. Vacuole sap proteins fraction-ated by gel filtration were studied by mass spectrometric analyses of trypsin and chymotrypsin digestions.
