This study aimed to establish culture medium changing system to improve the efficiency of bovine somatic cell nuclear transfer protocol. Our results showed that using the culture medium of Takahashi and First 1992 produced the cloned bovine blastocyst at the highest rate (9.4%, P < 0.05) among different modified synthetic oviduct fluid (mSOF) media.

The aim of the present study was to evaluate the effect of foetal calf serum (FCS) on the production of bovine embryo in vitro. We conducted two experiments: (1) to evaluate the effect of FCS on the production of bovine embryo in vitro and (2) to evaluate the effect of duration of FCS supplementation on the production of bovine embryo in vitro.

This study examined the effects of the cumulus cell co-culture system on the development of cloned bovine embryos. The results showed that the coculture of cloned bovine embryos with cumulus cells significantly improved the in vitro development rate to the blastocyst stage (25 %) compared to the control group (12.5 %).

Production of cloned bovine has many practical applications in medicine and agriculture, although low efficiency has still been the main problem. This challenge has resulted from improper epigenetic reprogramming of donor's nuclear such as histone acetylation and zygotic gene activation that does not occur correctly.

In this study, we evaluated the effects of corpus luteum (CL) size of local Vietnamese cattle and determined the ideally synchronized timing for cloned bovine embryos transfer. The results showed that there was a correlation between the size of corpus luteum of recipient cows and pregnancy rates after embryos transfer, in which the qualified recipient cow should consists of at least 1.7 cm in CL size at the time of transferring.

Animal cloning is the process of producing an entire organism that theoretically has the same genome as its donor cell line. To validate the success of SCNT, one must verify that the genetic source of the cloned organism is identical to the donor cell. However, none of the studies have performed to identify the genetic characteristics in the stage of preimplantation development of cloned embryo.

This technique is commercially expanding due to many improvements in embryo technology. In this study, we attempted to reduce the embryonic invasiveness and develop a simple, fast, and reliable sexing procedure using direct amplification of Y chromosome-specific marker from only one blastomere.

Assisted reproductive technologies (ART) are widely used to treat fertility issues in humans and for the production of embryos in mammalian livestock. The use of these techniques, however, is not without consequence as they are often associated with inauspicious pre- and postnatal outcomes including premature birth, intrauterine growth restriction and increased incidence of epigenetic disorders in human and large offspring syndrome in cattle.

Aberrant DNA methylation patterns of genes required for development are common in in vitro produced embryos. In this regard, we previously identified altered DNA methylation patterns of in vivo developed blastocysts from embryos which spent different stages of development in vitro, indicating carryover effects of suboptimal culture conditions on epigenetic signatures of preimplantation embryos.

Lactation and associated metabolic stresses during the post-partum period have been shown to impair fertility in dairy cows. The oviduct plays key roles in embryo development and the establishment of pregnancy in cattle.

Morphological evaluation of embryos has been used to screen embryos for transfer. However, the repeatability and accuracy of this method remains low. Thus, evaluation of an embryo’s gene expression signature with respect to its developmental capacity could provide new opportunities for embryo selection.

The homologous recombination (HR) pathway is largely inactive in early embryos prior to the first cell division, making it difficult to achieve targeted gene knock-ins. The homology-mediated end joining (HMEJ)-based strategy has been shown to increase knock-in efficiency relative to HR, non-homologous end joining (NHEJ), and microhomology-mediated end joining (MMEJ) strategies in non-dividing cells.

In this research, we have compared the growing capacity after thawing out frozen embryos which had different qualities and were formed from different source of oocytes. The results show that, the embryos which have standard forms, embryonic cells connected closely, without or few seperated cells or crushed pieces outside would have high growing capacity of frozen embryos after thawing out.
