Tensegrity is a structural system constructed of tensile cables and compressive bars. This thesis proposes several general methods for creating tensegrity structures. The author has developed new methods for creating specific tensegrity cells and the corresponding computer code. Furthermore, the author has constructed various physical models to validate the methods. A proposal for an observation tower on top of the Eureka building in Melbourne has also been devised by the author.

FTIR spectroscopy has been applied to study the coordination structures of Mg2þ and Ca2þ ions bound in Akazara scallop troponin C (TnC), which contains only a single Ca2þ binding site. The region of the COO – antisymmetric stretch provides information about the coordination modes of COO – groups to the metal ions: bidentate, unidentate, or pseudo-bridging. Two bands were observed at 1584 and 1567 cm21 in the apo state, whereas additional bands were observed at 1543 and 1601 cm21 in the Ca2þ-bound and Mg2þ-bound states, respectively.

The cloned guinea-pig Y2 neuropeptide Y (NPY) receptors expressed in Chinese hamster ovary (CHO) cells, as well as the Y2 receptors natively expressed in rat forebrain, are distributed in two populations. A smaller population that is readily accessed by agonist peptides on the surface of intact cells constitutes less than 30% of Y2 receptors detected in particulates after cell homogenization. A much larger fraction of cell surface Y2 sites can be activated by sulfhydryl modifiers.

We report the isolation of a novel antimicrobial peptide, bass hepcidin, from the gill of hybrid striped bass, white bass (Morone chrysops) · striped bass (M. saxatilis). After the intraperitoneal injection of Micrococcus luteus and Escherichia coli, the peptide was purified from HPLC fractions with antimicrobial activity against Escherichia coli. Sequencing by Edman degradation revealed a 21-residue peptide (GCRFCCNCCPNMSGCGVCCRF) with eight putative cysteines.

The solution structure of a synthetic mutant type I antifreeze protein (AFP I) was determined in aqueous solution at pH 7.0 using nuclear magnetic resonance (NMR) spectroscopy. The mutations comprised the replacement of the four Thr residues by Val and the introduction of two additional Lys-Glu salt bridges. The antifreeze activity of this mutant peptide, VVVV2KE, has been previously shown to be similar to that of the wild type protein, HPLC6 (defined here as TTTT).

We previously purified and characterized a peptide toxin, birtoxin, from the South African scorpion Parabuthus transvaalicus. Birtoxin is a 58-residue, long chain neurotoxin that has a unique three disulfide-bridged structure. Here we report the isolation and characterization of ikitoxin, a pep-tide toxin with a single residue difference, and a markedly reducedbiological activity, frombirtoxin. The chaperone calreticulin is a highly conserved eukaryotic protein mainly located in the endoplasmic reticulum. It contains a free cysteine SH group but does not form disul-fide-bridged dimers under physiological conditions,indica-ting that the SH group may not be fully accessible in the native protein. Using PAGE,urea gradient gel electro-phoresis,capillary electrophoresis and MS,we show that dimerization through the SH group can be induced by lowering the pH to 5–6,heating,or under conditions that favour partial unfolding such as urea concentrations above 2.

Pi4 is a38-residue toxincross-linkedby fourdisulfidebridges that has been isolated from the venom of the Chactidae scorpion Pandinus imperator. Together with maurotoxin, Pi1, Pi7 andHsTx1, Pi4belongs to theaKTX6 subfamilyof short four-disulfide-bridged scorpion toxins acting on K + channels. Due to its very low abundance in venom, Pi4 was chemically synthesized in order to better characterize its pharmacology and structural properties.

Neutralizing and protective monoclonal antibodies (mAbs) were used to fine-map the highlyconserved hemagglutinin noose epitope (H379–410, HNE) of themeasles virus. Short peptides mimicking this epitope were previouslyshown to induce virus-neutralizing antibodies [El Kasmi et al. (2000) J. Gen. Virol.81, 729–735]. The epitope contains three cys-teine residues, two of which (Cys386 and Cys394) form a disulfide bridge critical for antibodybinding.

To elucidate the structural features of the mussel defensin MGD1 required for antimicrobial activity, we synthesized a series of peptides corresponding to the main known secon-dary structures of the molecule and evaluated their activity towards Gram-positive and Gram-negative bacteria, and filamentous fungi. We found that the nonapeptide corres-ponding to residues 25–33 of MGD1 (CGGWHRLRC) exhibited bacteriostatic activity once it was cyclized by a non-naturally occurring disulfide bridge.
