A bifunctional glycoside hydrolase GH78 from the ascomycete Xylaria polymorpha (XpoGH78) with catalytic versatility to both glycosides and esters, may be beneficial for the efficient degradation of plant cell-wall complexes containing both diverse sugar residues and esterified structures to apply in, for example, the production of biofuels from plant feedstock.

Feruloyl esterase catalyze the hydrolysis of a wide variety of glycosidic linkages between two or more carbohydrates or between a carbohydrate and a non-carbohydrate moiety. Thereby, they play a role in the initial degradation of lignocelluloses, which may be advantageous for the efficient degradation of the plant cell-wall complex that contains both diverse sugar residues and esterified structures.

Relatively little is known about the genomic basis and evolution of wood-feeding in beetles. We undertook genome sequencing and annotation, gene expression assays, studies of plant cell wall degrading enzymes, and other functional and comparative studies of the Asian longhorned beetle, Anoplophora glabripennis, a globally significant invasive species capable of inflicting severe feeding damage on many important tree species.

Leaf blight caused by Calonectria spp. is one of the most destructive diseases to affect Eucalyptus nurseries and plantations. These pathogens mainly attack Eucalyptus, a tree with a diversity of secondary metabolites employed as defense-related phytoalexins. To unravel the fungal adaptive mechanisms to various phytoalexins, we examined the genome of C. pseudoreteaudii, which is one of the most aggressive pathogens in southeast Asia.

Pectobacterium parmentieri is a newly established species within the plant pathogenic family Pectobacteriaceae. Bacteria belonging to this species are causative agents of diseases in economically important crops (e.g. potato) in a wide range of different environmental conditions, encountered in Europe, North America, Africa, and New Zealand. Severe disease symptoms result from the activity of P. parmentieri virulence factors, such as plant cell wall degrading enzymes.

Pectin is a major component and structural polysaccharide of the primary cell walls and middle lamella of higher plants. Pectate lyase (PEL, EC 4.2.2.2), a cell wall modification enzyme, degrades de-esterified pectin for cell wall loosening, remodeling and rearrangement. Nevertheless, there have been few studies on PEL genes and no comprehensive analysis of the PEL gene family in cotton.

Lentinula edodes is one of the most popular edible mushroom species in the world and contains useful medicinal components, such as lentinan. The light-induced formation of brown film on the vegetative mycelial tissues of L. edodes is an important process for ensuring the quantity and quality of this edible mushroom.

Pectobacterium carotovorum subsp. brasiliense is a broad host range bacterial pathogen, which causes blackleg of potatoes and bacterial soft rot of vegetables worldwide. Production of plant cell wall degrading enzymes is usually critical for Pectobacterium infection. However, other virulence factors and the mechanisms of genetic adaptation still need to be studied in detail.

The genetic diversity in Australian populations of Xanthomonas species associated with bacterial leaf spot in tomato, capsicum and chilli were compared to worldwide bacterial populations. The aim of this study was to confirm the identities of these Australian Xanthomonas species and classify them in comparison to overseas isolates.

Studies on biochemical changes (soluble sugar, phenol, protein, ash, phosphorus, calcium) and changes in cell wall degrading enzymes (PG, PMG and CX) in papaya fruit inoculated with Colletotrichum demetium was carried out. Total protein content of inoculated papaya fruits was decreased by 0.63, 0.58 and 0.36 per cent at 6th, 7th and 8th day after inoculation, respectively, while in control fruit protein content was 1.80 mg/g. The TSS content in inoculated papaya fruits is progressively decreased by 3.93, 3.71 and 3.42 per cent at 6th , 7 th and 8th day after inoculation.

Cellulose constitutes bulk of the plant cell wall materials and the most abundant and renewable non-fossil carbon source on earth. Cellulose degrading fungi play an important role in recycling of cellulosic materials. Thus, this study was designed to screen endophytic fungi isolated from Cameroonian medicinal plants for their cellulase activity. The plate-clearing assay and enzyme assay methods were used for screening of potent cellulolytic endophytic fungi. The promising was chosen for filter paper cellulase (FPCase) and carboxymethyl cellulase (CMCase) assay.

In order to extend the shelf life of banana cultivar Tella Chakkerakeli was fumigated with 1-methylcyclopropane (1-MCP) at four different concentrations (100, 200, 300 and 400 ppb) @ 24h exposed time and stored at ambient conditions. Among all the treatments, 400 ppb was found to be an effective in extending shelf-life, highest retention of cell wall degrading enzyme such as pectin methylestarase and antioxidant enzymes, peroxidase and catalase. The entire 1-MCP treated banana showed better results over control (kept at open air @ 24h).

The plant cell wall serves as a primary barrier against pathogen invasion. The success of a plant pathogen largely depends on its ability to overcome this barrier. During the infection process, plant parasitic nematodes secrete cell wall degrading enzymes (CWDEs) apart from piercing with their stylet, a sharp and hard mouthpart used for successful infection.

Sugarcane is a tropical crop that can accumulate high concentration of sucrose in the stem as a storage carbohydrate. For that reason, sugarcane accounts for approximately 75% of all the sugar produced in the world and has become the main sugar source to produce first-generation bioethanol in Brazil.
