As a first step toward developing a zebrafish model for investigating the role of sulfation in counteracting environ-mental estrogenic chemicals, we have embarked on the identification and characterization of cytosolic sulfotrans-ferases (STs) in zebrafish. By searching the zebrafish expressed sequence tag database, we have identified two cDNA clones encoding putative cytosolic STs. These two zebrafish ST cDNAs were isolated and subjected to nuc-leotide sequencing. Sequence data revealed that the two zebrafishSTs are highlyhomologous, being

The cloning of two novel genes that encode cytokinin oxidase/dehydrogenase (CKX) in barley is described in this work. Transformation of both genes intoArabidopsisand tobacco showed that at least one of the genes codes for a functional enzyme, as its expression caused a cytokinin-deficient phenotype in the heterologous host plants. Addi-tional cloning of two gene fragments, and anin silicosearch in the public expressed sequence tag clone databases, revealed the presence of at least 13 more members of the CKXgene family in barley and wheat. ...

A novel cytochrome P450, CYP4x1, was identified in EST databases on the basis of similarity to a conserved region in the C-helix of the CYP4A family. The human and mouse CYP4x1 cDNAs were cloned and found to encode putative cytochrome P450 proteins. Molecular modelling of CYP4x1 predicted an unusual substrate binding channel for the CYP4 fam-ily.

Despite its importance in plant metabolism, no sequences of higher plant ATP-dependent phosphofructokinase (EC 2.7.1.11) are annotated in the databases. We have purified the enzyme from spinach leaves 309-fold to electrophoretic homogeneity. The purified enzyme was a homotetramer of 52 kDa subunits with a specific activity of 600 mUÆmg )1 and aKmvalue for ATP of 81 lm.

Four cDNA clones that were annotated in the database as encoding d-amino acid oxidase (DAAO) or d-aspartate oxidase (DASPO) were iso-lated by RT-PCR from Caenorhabditis elegans RNA. The proteins (Y69Ap, C47Ap, F18Ep, and F20Hp) encoded by the cloned cDNAs were expressed in Escherichia colias recombinant proteins with an N-terminal His-tag.
