The present investigation was carried out in Biotechnology Laboratory, Department of Olericulture, KAU, Vellanikkara with four genotypes of cucumber comprising gynoecious, monoecious and two parthenocarpic genotypes for in vitro plant regeneration. Seeds were grown in the culture bottles quarterly filled with half strength MS basal medium. Cotyledonary leaf explants were then transferred to eight media compositions of half strength MS medium with varying IAA and BAP concentrations in the culture test tubes.

The present study was carried out during Late kharif, 2016 at College of Horticulture, Anantharajupeta. The experiment was conducted with an objective to identify divergent genotypes to be used as donor parents in crop improvement. Thirty-two genotypes along with two checks were evaluated in a randomized block design with three replications and the data on 25 yield and yield attributing traits were recorded. The analysis of variance revealed significant differences among all genotypes indicating the presence of sufficient amount of variability for the characters studied.

The present experiment was carried out to assess the degree of association of yield with its components, because yield is not an independent character and it is resultant of interaction of a number of component characters. Correlation provides information on the nature and extent of association between characters in a population. Most of the traits have shown significant correlation as revealed by the association study.

Thirteen (Three gynoecious and ten monoecious) germplasm lines of cucumber (Cucumber sativus L.) were characterized by sodium dodecyl sulphate polyacrylamide vertical slab gel electrophoresis (SDS-PAGE). The seed protein could be resolved into total 11 bands distributed in 4 zones i.e. A, B, C and D. Zone A was divided into 5 subzones and 5 bands, zone B has 1 band C has 5 and zone D included 5 bands. Similarity index value ranged from 62% to 100% among all the genotypes. Pgyn-1 showed least similarity 68% with other genotypes.

A field investigation was carried out with thirty genotypes of cucumber to evaluate the performance of the genotypes for growth and yield characters under hill zone of Karnataka. The genotype Sirsi Local-2 exhibited superiority for vine length, number of primary branches per plant, days to first harvest of the fruit, per cent marketable fruits per vine and chlorophyll content. Minimum number of node to appear first female flower, highest number of fruits per vine (13.90) and maximum yield per vine were found in the cultivar Poinsette.

Fifty-six diverse genotypes of cucumber collected from different indigenous sources were planted in randomized block design, during kharif season at Kittur Rani Chennamma College of Horticulture, Arabhavi, Belagavi district (Karnataka), India. Genetic diversity study using Mahalanobis’s D2 statistics with Tocher’s method, 56 cucumber genotypes were grouped into ten clusters, cluster I was the largest having 26 genotypes followed by cluster III (10 genotypes) followed by cluster II (six genotypes).

With the objective to find the association of different seed characters with fruit yield in cucumber, an experiment was conducted with 30 indigenous genotypes, collected from five different states, at the Experimental Farm of the Department of Vegetable Science, Dr YS Parmar University of Horticulture and Forestry, Nauni, Solan during Kharif, 2015. Results revealed that yield per plant had positive and significant association with all the seed characters under study viz., seed length, seed breadth, hundred seed weight, germination percentage, seed vigour index-I and seed vigour index-II.

Evaluation of 19 diverse genotypes of cucumber was carried out in a randomized block design for studying genetic divergence. Presence of wide genetic diversity, among the 19 genotypes studied was confirmed by using the powerful tool of Mahalanobis D2 statistic. The present investigation were carried out understand heterosis and gene action for yield and quality traits in cucumber genotypes. The materials consist of three lines and four testers and the resultant 12 hybrids through Line x Tester mating design. The present estimation of heterosis identified that L2XT2 (Koradacherry local x Orathanadu local) being the superior hybrid for most of the traits by expressing higher significance Relative Heterosis, Heterobeltiosis and Standard Heterosis.

Fifteen gynoecious parthenocarpic hybrids hybrids/lines of cucumber (Cucumis sativus L.) were evaluated under polyhouse (fifty percent shade/naturally ventilated) conditions for yield and quality. The genotypic and phenotypic coefficient of variations were highest for nodal position of first female flower, number fruits, fruit weight and fruit yield per plant which indicates maximum variability is existing in the genotypes. High genetic advance as per cent mean was observed for most of the characters, indicating that these characters are controlled by additive gene action.
