Several commercial and freely-available tools can be used to analyze DNA fingerprint gel images; however, commercial tools are expensive and usually difficult to use; and, free tools support the basic functionality for DNA fingerprint analysis, but lack some instrumental features to obtain accurate results.

The reference marker set (30 markers) was used to evaluate the genetic diversity and DNA fingerprinting of 19 standard reference rice varieties. The results showed the similarity coefficient of 19 varieties varied from 0.04 to 0.548. At the genetic similarity coefficient of 0.1, the 19 rice varieties divided into two main groups. Group one included 3 varieties: DH1, DH5, DH13. Group 2 included the remaining 16 varieties. Inside group two, phylogenetic tree divided into two main branches at the genetic similarity coefficient of 0.3.

Soybean is legume crop having high protein (40%) and oil (20%) content. It has highest share in production among all oilseeds of the world. In present study 10 soybean genotypes were characterized using 14 RAPD primers. Total 103 amplicons were obtained out of which 75 were polymorphic and 28 were monomorphic. The percent polymorphism obtained was 72.81%. Highest average similarity coefficient was exhibited by the genotype MAUS-32 (0.675) and lowest similarity coefficient exhibited by MAUS-2 (0.584). The dendrogram of these genotypes grouped into two main clusters.

A set of 45 polymorphic microsatellite markers, representing markers for almost each chromosome, was used for the assessment of genetic diversity in 35 genotypes of bread wheat. A total of 127 alleles were detected which ranged from 2 to 6 with an average allele number of 2.82 per locus. The PIC value of the 45 SSRs ranged from 0.08 to 0.50 with an average PIC of 0.30. Resolving power and Marker index of all 45 markers varied from 0.17 to 2.82 and 0.16 to 1.52, respectively.

Twenty three simple sequence repeat markers were used to study genetic relationship among fifty three rice varieties developed from Andhra Pradesh and Telangana states. All markers were polymorphic among 53 varieties. A total of 55 alleles were produced for 23 markers with fragment size ranging from 60bp (RM204) to 470bp (RGNMS1289). The number of alleles per marker were ranged from two (RM262) to four (RGNMS1776) with an average of 2.39 alleles/loci.

Random amplified polymorphism DNA (RAPD) analysis was done to assess the diversity among 21 species/strains of Pleurotus. The morphologically similar species/ strains too gave a new account of the evolutionary process and taxonomy of mushrooms. A total of 150 10 mer primers were screened, out of which 10 primers viz. OPD-03, OPD-05, OPD08, OPA-13, OPA-16, OPQ-15, OPQ-16 and OPQ-18, S-1461 and S-1462 produced consistent polymorphic banding pattern.

In this study, RAPD fingerprinting has been carried out for variants selected from orchards growing with micropropagated Robusta banana (Musa spp. ‘AAA’) plants. After isolation of quality DNA and preliminary screening of random primers, fourteen arbitrary (10-mers) primers showing intense unambiguous and reproducible PCR amplification have been selected for study. Among them OPA-19 and OPC-03 showed only monomorphic bands while OPA-02, OPA-09, OPA-13, OPA-14, OPB-6, OPB-15, OPC-01, OPD-10, OPF-04 and OPF-12 primers showed polymorphic bands.
