Krüppel-type zinc finger genes (ZNF) constitute a large yet relatively poorly characterized gene family. ZNF genes encode proteins that recognize specific DNA motifs in gene promotors. They act as transcriptional co-activators or -repressors via interaction with chromatin remodeling proteins and other transcription factors.

Here we show that Eos associates with the corepressor C-terminal-binding protein (CtBP). CtBP has previously been shown to bind Pro-X-Asp-Leu-Ser (PXDLS) motifs in several DNA-binding proteins. Tetracycline repressor (TetR), which constitutes the most commonmechanismof bacterial resistance to an antibiotic, is a homodimeric protein composed of two identical sub-units, each of which contains a domain possessing a helix– turn–helix motif and a domain responsible for binding tetra-cycline. Binding of tetracycline in the protein pocket is accompanied by conformational changes in TetR, which abolish the specific interaction between the protein and DNA.

Human MOK2 is a DNA-binding transcriptional repressor. Previously, we identified nuclear lamin A⁄C proteins as protein partners of hsMOK2. Fur-thermore, we found that a fraction of hsMOK2 protein was associated with the nuclear matrix. We therefore suggested that hsMOK2 interactions with lamin A⁄C and the nuclear matrix may be important for its ability to repress transcription.
