Mitochondrial DNA (mtDNA) deletion mutations accumulate with age in tissues of a variety of species. Although the relatively low calculated abundance of these deletion mutations in whole tissue homogenates led some investigators to suggest that these mutations do not have any physiological impact, their focal and segmental accumulation suggests that they can, and do, accumulate to levels sufficient to affect the metabolism of a tissue.

In the O2– generating flavocytochrome b, the membranebound component of the neutrophil NADPH oxidase, electrons are transported from NADPH to O2 in the following sequence: NADPH fi FAD fi heme b fi O2. Although p-iodonitrotetrazolium (INT) has frequently been used as a probe of the diaphorase activity of the neutrophil flavocytochrome b, the propensity of its radical to interact reversibly with O2 led us to question its specificity. This study was undertaken to reexamine the interaction of INT with the redox components of the neutrophil flavocytochrome b. ...

Hydrogenase and fumarate reductase isolated fromWoli-nella succinogeneswere incorporated into liposomes con-taining menaquinone. The two enzymes were found to be oriented solely to the outside of the resulting proteolipo-somes. The proteoliposomes catalyzed fumarate reduction by H2 which generated an electrical proton potential (Dw 0.19 V, negative inside) in the same direction as that gen-erated by fumarate respiration in cells ofW. succinogenes.

Complex I (NADH: ubiquinone oxidoreductase) is the first complex in the respiratory electron transport chain. Homologs of this complex exist in bacteria, mitochondria and chloroplasts. The minimal complex I from mitochondria and bacteria contains 14 different subunits grouped into three modules: membrane, connecting, and soluble subcomplexes. The com-plex I homolog (NADH dehydrogenase or Ndh complex) from chloroplasts from higher plants contains genes for two out of three modules: the mem-brane and connecting subcomplexes. ...

A highly purified cytochromeb6f complex from the cyanobacterium Syn-echocystissp. PCC 6803 selectively binds one chlorophyll a and one caro-tenoid in analogy to the recent published structure from two other b6 f complexes. The unknown function of these pigments was elucidated by spectroscopy and site-directed mutagenesis. Low-temperature redox differ-ence spectroscopy showed red shifts in the chlorophyll and carotenoid spec-tra upon reduction of cytochrome b6, which indicates coupling of these pigments with the heme groups and thereby with the electron transport....

Transport of presecretory proteins into the mammalian rough endoplasmic reticulum involves a protein translocase that comprises the integral membrane proteins Sec61ap, Sec61bp, and Sec61cp as core components. Electron microscopic analysis of protein translocase in rough micro-somal membranes suggested that between three and four heterotrimeric Sec61p complexes form the central unit of protein translocase.

The His332 residue of the D1 protein has been identified as the likely ligand of the catalytic Mn ions in the water oxi-dizing complex (Ferreira, K.N., Iverson, T.M., Maghlaoui, K., Barber, J. & Iwata, S. (2004) Science303, 1831–1838). However, its function has not been fully clarified. Here we used thermoluminescence and flash-induced chlorophyll fluorescence measurements to characterize the effect of theD1-H333E,D1-H332DandD1-H332Smutationson the electron transport of Photosystem II in intact cells of the cyanobacteriumSynechocystis6803....

Melithiazol and myxothiazol are two myxobacterial metabolites that are highly efficient electron transport inhibitors of the respiratory chain. MelJ and MelK encoded in the melithiazol biosynthetic gene cluster were recently shown to be involved in the formation of the methyl ester from a hypothetical amide intermediate.

Subunit 4 of cytochromecoxidase (CcO) is a nuclear-encoded regulatory subunit of the terminal complex of the mitochondrial electron transport chain. We have recently discovered an isoform of CcO 4 (CcO4-2) which is specific to lung and trachea, and is induced after birth. The role of CcO as the major cellular oxygen consumer, and the lung-specific expression of CcO4-2, led us to investigate CcO4-2gene regulation.

The chloroplast thylakoid is the most abundant membrane system in nat-ure, and is responsible for the critical processes of light capture, electron transport and photophosphorylation. Most of the resident proteins are imported from the cytosol and then transported into or across the thyla-koid membrane.

In the apicoplast of apicomplexan parasites, plastidic-type ferredoxin and ferredoxin-NADP + reductase (FNR) form a short electron transport chain that provides reducing power for the synthesis of isoprenoid precursors. These proteins are attractive targets for the development of novel drugs against diseases such as malaria, toxoplasmosis, and coccidiosis.

Electron transfer proteins transport electrons safely between large redox enzymes. The complexes formed by these proteins are among the most transient. The biological function requires, on the one hand, sufficient spec-ificity of the interaction to allow for rapid and selective electron transfer, and, on the other hand, a fast turnover of the complex.
