We have isolated and studied the cloned sigma factors SASIG1-3 from mustard (Sinapis alba). In functional ana-lyses using both promoter and factor mutants,the three recombinant proteins all had similar basic properties but also revealed differences in promoter preference and requirements for single nucleotide positions. Directedmuta-genesis of SASIG1 identified critical residues within the conserved regions 2.4 and 4.2 necessary for binding of the )10 and)35 promoter elements,respectively.

Tobacco pathogenesis-related protein 1a (PR-1a) is induced in plants during the hypersensitive response (HR) after exposure of plants to salicylic acid (SA) and by develop-mental cues.Gene activation by these diverse stimuli is mediated via anas-1-like element in the PR-1aupstream region.To further analyze the significance of thiscis-acting sequence, an authentic as-1element from the cauliflower mosaic virus 35S RNA promoter was inserted into the PR-1apromoter in place of theas-1-like motif.

The TPaand TPbisoforms of the human thromboxane A2 receptor (TP) arise by differential splicing but are under the transcriptional control of two distinct promoters, termed Prm1 and Prm3, respectively (Coyle et al. 2002Eur J Biochem269, 4058–4073). The aim of the current study was to determine the key factors regulating TPbexpression by functionally charac-terizing Prm3, identifying the core promoter and the cis-acting elements regulating basal Prm3 activity.

Lactococcus lactisFI9078, a construct carrying a disruption of theldh gene, converted approximately 90% of glucose into lactic acid, like the parental strain MG1363. This unexpected lactate dehydrogenase activity was puri-fied, andldhB was identified as the gene encoding this protein. The basic distinction between already known algorithmic characterizations of matroids and antimatroids is in the fact that for antimatroids the ordering of elements is of great importance. While antimatroids can also be characterized as set systems, the question whether there is an algorithmic description of antimatroids in terms of sets and set functions was open for some period of time.
