The interaction of bacterial endotoxins [lipopolysaccharide (LPS) and theendotoxic principle lipid A], with high-den-sity lipoprotein (HDL) from serum was investigated with a variety of physical techniques and biological assays. The folding of a 93-residue protein, the histidine-phospho-carrier protein ofStreptomyces coelicolor,HPr,hasbeen studied using several biophysical techniques, namely fluo-rescence, 8-anilinonaphthalene-1-sulfate binding, circular dichroism, Fourier transform infrared spectroscopy, gel filtration chromatography and differential scanning calori-metry. AP1 (GEQGALAQFGEWL) was shown by theoretical analysis to be an anionic oblique-orientateda-helix former. The peptide exhibited a mono-layer surface area of 1.42 nm 2 , implying possession of a-helical structure at an air⁄water interface, and Fourier transform infrared spectroscopy (FTIR) showed the peptide to be a-helical (100%) in the presence of vesi-cle mimics of Escherichia colimembranes.
