Gamma rays and EMS

In this study, hard wood cuttings of Hibiscus rosa-sinenis Cultivar Red Single were treated with nine different dose of gamma rays (1 to 5 k R) and ten different concentrations of EMS ranging from 0.1% to 1.0%. The results revealed that gradual reduction in survival of cuttings, shoot length, leaf length and leaf width with increase in dosage of gamma rays and EMS. However gamma rays treated plants showed significant gradual reduction in growth rate than the EMS treated plants. Based on probit analysis, LD50 (Lethal dose) value for gamma radiation and EMS have been fixed as 2.81 kR and 0.

The biological effects of physical mutagen gamma rays and chemical mutagen Ethyl Methyl Sulphonate (EMS) were analyzed in two varieties of cowpea viz., VBN-1, and C152. Five different doses of gamma rays (100 Gy, 200 Gy, 300 Gy, 400 Gy and 500 Gy) and five different concentrations of EMS (5 mM, 10 mM, 15 mM, 20 mM and 25 mM) were used for inducing mutations in these two varieties.

The current study was conducted to find out the LD50 dose, shoot length and root length induced by both gamma rays and EMS in blackgram variety VBN 4, under laboratory condition. The experiment was done with two mutagenic agents viz., Gamma radiation (γ) and EMS and two varieties namely BCO-1 and Japanese Jhar Bhendi. Seeds were irradiated with gamma rays at 100, 200, 300, 400, 500 kRγ and treated with 0.1, 0.2, 0.3, 0.4 and 0.5 per cent EMS concentration. A total of 10gamma irradiated lines and 10 EMS treated lines along with dry and wet controls of the two varieties were grown in M2 generation. The first two principle components having Eigen values more than one were cumulatively contributing 99.57% to the total induced variation.

An experiment was conducted to study the effectiveness and efficiency of one physical mutagen i.e. gamma rays and two chemical mutagens i.e. ethyl methanesulfonate (EMS) and nitrosoguanidine (NG) in greengram. An investigation was carried out at the Department of Floriculture and Landscaping, Horticultural College and Research Institute, Tamil Nadu Agricultural University, Coimbatore during 2014-2016 on the improvement of tuberose var. Prajwal through mutation breeding. The bulbs were treated with gamma rays, diethyl sulphate (DES) and ethyl methane sulphonate (EMS). The treatments consisted of 0.5, 1.0, 1.5, 2.0 and 2.5 kR of gamma rays, 15, 20, 25 and 30 mM of DES and 30, 45, 60 and 75 mM of EMS and control (untreated). Various morphological and floral characters were observed.

Field trials were conducted to study the effect of physical and chemical mutagens (Gamma rays, Ethyl Methane Sulphonate (EMS) and Diethyl sulphate (DES)) on biometric characters viz., days to 50% flowering, plant height, number of branches, fruits per plant, fruit length, fruit girth, fruit weight and fruit yield per plant and their correlations in five brinjal cultivars viz., Angoor, Annamalai, Hissar pragath, PLR 1 and Putheri. The results revealed that there had been strong association between number of fruits per plant and fruit weight with fruit yield per plant in M2 generation.

A systematic and comparative study on mutagenic ability of different doses of ethyl methane sulphonate (EMS, an alkylating agent), gamma rays (an ionizing radiation) and EMS + gamma rays was carried out in a well-adapted desi chickpea variety HPG17. All mutagenic treatments were effective in inducing genetic variability. A proportional decrease in germination of mutagen treated seeds as well as subsequent plant survival was observed with increase in mutagen dose.

Induced qualitative mutation in JS-335 Soybean cultivar consisted of chlorophyll and viable mutants. The combination treatments exhibit wide spectrum and frequency on both M1 family and M2 plant basis for both. Increased in doses and concentrations of mutagens showed increased in spectrum and frequency. Chlorophyll mutants viz. albino, alboviridis, xantha, xantha- viridis and viridis and viable mutants viz., plant type, leaf, growth habit, flowering, pod, kernel and economic mutation were observed. Desirable mutants like bold poded, early maturing, and high yielding were also isolated.

The present investigation was conducted during the Kharif season of 2013, seeds of black gram [Vingo mungo (L.) Hepper] variety VBN 4 and ACM 07002 were exposed to gamma rays doses at 200, 250, 300 and 350 Gy and EMS (Ethyl Methane Sulphonate) doses at 20, 30 and 40 mM. Wide ranges of viable mutants were isolated in M2 and M3 generation. The mutational change in each quantitative trait can be large or small.

The present investigation was carried out to find the efficiency and effectiveness of various mutagenic treatments of gamma rays and EMS on cape gooseberry. Genetically pure, uniform and dry seeds of cape gooseberry were subjected to 2, 5, 10, 15 and 20 kR doses of gamma rays and 0.02, 0.03, 0.04 and 0.05 M concentration of ethyl methane sulfonate along with control. Seed treatment with gamma rays, EMS, NG and their combinations in two mungbean genotypes, BKG-1 and OUM 11-5, significantly reduced germination, seedling growth including fresh weight and dry weight, pollen fertility, seed fertility and survival at maturity in M1 over the parents. The biological damage in M1 generation showed a dose dependent linear relationship. NG in both single and combination treatments resulted in more pronounced biological damage than other treatments.

Seed protein content did not show any significant positive correlation with seed size and test weight. Heritability estimates were quite high for almost all polygenic traits among the mutants, in comparison to the control. Cytological analysis of the mutants was almost normal, except for a few chromosomal aberrations.

Combination treatments were most effective in inducing a wider spectrum and maximum frequency of macro mutations, followed by EMS. Of all the morphological mutations, the frequency of leaf mutations was maximum, followed by plant height and seed mutants.
