It is a basic task in high-throughput gene expression profiling studies to identify differentially expressed genes (DEGs) between two phenotypes. RankComp, an algorithm, could analyze the highly stable withinsample relative expression orderings (REOs) of gene pairs in a particular type of human normal tissue that are widely reversed in the cancer condition, thereby detecting DEGs for individual disease samples measured by a particular platform.

In order compute genetic variability, heritability and genetic advance expressed as per cent of mean, an investigation was carried out with 79 plants of F2 population of MFC-09-12 × PGCP-12 and 90 plants of F2population of MFC-08-14 × PL-3 in cowpea. Considerably high amount of variability was observed for the thirteen quantitative and qualitative characters under study.

Comparative analyses of chloroplast and mitochondrial genomes have shown that organelle genomes in bryophytes evolve slowly. However, in contrast to seed plants, the organellar genomes are yet poorly explored in bryophytes, especially among liverworts.

The human b-globin gene locus is the subject of intense study, and over the past two decades a wealth of information has accumulated on how tissue-specific and stage-specific expression of its genes is achieved. The data are extensive and it would be difficult, if not impossible, to formulate a comprehensive model integrating every aspect of what is currently known. In this review, we introduce the fundamental characteristics of globin locus regulation as well as questions on which much of the current research is predicated.

The gene fprA of Mycobacterium tuberculosis, encoding a putative protein with 40% identity to mammalian adrenodoxin reductase, was expressed in Escherichia coli and the protein purified to homogeneity. The 50-kDa protein monomer contained one tightly bound FAD, whose fluorescence was fully quenched. FprA showed a low ferric reductase activity, whereas it was very active as a NAD(P)H diaphorase with dyes. Kinetic parameters were determined and the specificity constant (kcat/Km) for NADPH was two orders of magnitude larger than that of NADH....

Mitochondrial uncoupling protein 1 (UCP1) is a speci®c marker of multilocular brown adipocytes. Ectopic UCP1 in white fat of aP2-Ucp1micemitigates development of obesity byboth, increasing energy expenditure anddecreasingin situ lipogenesis. In order to further analyse consequences of respiratory uncoupling inwhite fat, the eects of the ectopic UCP1 on the morphology of adipocytes and biogenesis of mitochondria in these cells were studied.

ThenblAfamily of genes encodes for small proteins neces-sary for the ordered degradation of phycobilisomes under certain stress conditions, a process known as chlorosis. Genes homologous tonblAseem to occur in all phycobili-some-containingorganisms.However, todate, nomolecular mechanism is known for the action of NblA, nor have the gene products been characterized to understand the physical properties of the molecule and thus help elucidate the mechanism on a structural basis.

The Escherichia coli bifunctional T-protein transforms chorismic acid to p-hydroxyphenylpyruvic acid in the L-tyrosine biosynthetic pathway. The 373 amino acid T-protein is a homodimer that exhibits chorismate mutase (CM) and prephenate dehydrogenase (PDH) activities, both of which are feedback-inhibited by tyrosine. Fifteen genes coding for the T-protein and various fragments thereof were constructed and successfully expressed in order to charac-terize theCM, PDHand regulatory domains.

PSA (prostate-specific antigen), the most useful serum marker for prostate cancer, is encoded by thehKLK3gene and is present in the serumas amixture of several molecular species. This work was performed to identify the hKLK3 transcripts in order to determine how many proteins resembling PSA are synthesized from thehKLK3gene and secreted in blood. Combined Northern blotting, molecular cloninganddatabase searching showed that thehKLK3gene produces at least 15 transcripts ranging in size from 0.7 to 6.1 kb. Polysomal distribution analysis revealed that the transcripts shorter than 3.

In order to study the role of peroxisome proliferator-acti-vated receptor ain mouse intestine, its agonist-induced proteins were identified by peptide mass fingerprinting fol-lowed byNorthern blot analysis using their cDNAs. One of the most remarkably induced proteins was identified as 17b-hydroxysterol dehydrogenase type 11. Its very rapid induction by various agonists was most efficient in intestine and then in liver.

Malonyl-CoA, a potent inhibitor of carnitine pamitoyl transferase-I (CPT-I), plays a pivotal role in fuel selection in cardiac muscle. Malonyl-CoA decarboxylase (MCD) cata-lyzes the degradation of malonyl-CoA, removes a potent allosteric inhibition on CPT-I and thereby increases fatty acid oxidation in the heart. Although MCDhas several Ser/ Thr phosphorylation sites, whether it is regulated by AMP-activated protein kinase (AMPK) has been controversial. We therefore overexpressed MCD(Ad.MCD) and consti-tutively active AMPK (Ad.

GenesSNO1 and SNZ1 are Saccharomyces cerevisiae homologues ofPDX2andPDX1which participate in pyri-doxine synthesis in the fungusCercospora nicotianae. In order to clarify their function, the two genesSNO1and SNZ1were expressed inEscherichia colieither individually or simultaneously and with or without a His-tag. When expressed simultaneously, the two protein products formed a complex and showed glutaminase activity.

TRPP2, also called polycystin-2, the gene product ofPKD2, is a membrane protein defective in 10–15% of cases of autosomal dominant polycystic kidney disease. Mutations in PKD2 are also associated with extrarenal dis-orders, such as hepatic cystogenesis and cardiovascular abnormalities.

The Drosophila melanogaster multisubstrate deoxyribonucleoside kinase (dNK; EC 2.7.1.145) has a high turnover rate and a wide substrate range that makes it a very good candidate for gene therapy. This concept is based on introducing a suicide gene into malignant cells in order to activate a prodrug that eventually may kill the cell.

Serial analysis of gene expression (SAGE) is a powerful technique for studying gene expression at the genome level. However, short SAGE tags limit the further study of related data. In this study, in order to identify a gene, we developed a semi-nested PCR-based method called the two-step analysis of unknown SAGE tags (TSAT-PCR) to generate longer 3¢-end cDNA fragments from unknown SAGE tags.

Mutations in the alanine-glyoxylate aminotransferase gene (AGXT) are responsible for primary hyperoxaluria type I, a rare disease characterized by excessive hepatic oxalate production that leads to renal failure. A deeper understanding of the changes in the metabolic pathways secondary to the lack of AGXT expression is needed in order to explore substrate depletion as a therapeutic strategy to limit oxalate production in primary hyperoxal-uria type I.
