Ethylene signal transduction in plants is conducted by the two-component system (TCS) which consists of histidine kinase (HK), histidine phosphotransferase (HPT) and response regulators (RRs).

Legumes can establish on nitrogen-deprived soils a symbiotic interaction with Rhizobia bacteria, leading to the formation of nitrogen-fixing root nodules. Cytokinin phytohormones are critical for triggering root cortical cell divisions at the onset of nodule initiation.

The fungicide fludioxonil over-stimulates the fungal response to osmotic stress, leading to overaccumulation of glycerol and hyphal swelling and bursting. Fludioxonil-resistant fungal strains that are null-mutants for osmotic stress response genes are easily generated through continual sub-culturing on sub-lethal fungicide doses.

Two component systems (TCS) are signalling complexes manifested by a histidine kinase (receptor) and a response regulator (effector). They are the most abundant signalling pathways in prokaryotes and control a wide range of biological processes.

The purified soluble forms of the histidine kinases BvgS and EvgS of Bordetella pertussis and Escherichia coli, respectively, are shown to be responsive to oxidized ubiquinone-0 (Q-0) in vitro. The oxidized ubiquinone is a strong inhibitor of kinase activity of both enzymes with half maximal inhibition occurring at 11 lM (BvgS) and 4 lM (EvgS). Reduced Q-0 has no effect on the histidine kinases. Kinase activity can reversibly be switched off and on by changing the oxidation status of the quinone....

Pyruvate dehydrogenase kinase (PDK) is the primary regulator of flux through the mitochondrial pyruvate dehy-drogenase complex (PDC).Analysis of the primary amino-acid sequences of PDK from various sources reveals that these enzymes include the five domains characteristic of prokaryotic two-component His-kinases, despite the fact that PDKexclusivelyphosphorylates Ser residues in theE1a subunit of the PDC.This seeming contradiction might be resolved if the PDK-catalyzed reaction employed a phos-pho-His intermediate....

In proteobacteria capable of H2oxidation under (micro)aerobic conditions, hydrogenase gene expression is often controlled in response to the availab-ility of H2. The H2 -sensing signal transduction pathway consists of a het-erodimeric regulatory [NiFe]-hydrogenase (RH), a histidine protein kinase and a response regulator.

The signal of the plant hormone cytokinin is perceived by membrane-located sensor histidine kinases and transduced by other members of the plant two-component system. InArabidopsis thaliana, 28 two-component system proteins (phosphotransmitters and response regulators) act down-stream of three receptors, transmitting the signal from the membrane to the nucleus and modulating the cellular response.

Bacteriophytochromes constitute a light-sensing subgroup of sensory kin-ases with a chromophore-binding motif in the N-terminal half and a C-ter-minally located histidine kinase activity. The cyanobacteriumFremyella diplosiphon(also designated Calothrixsp.) expresses two sequentially very similar bacteriophytochromes, cyanobacterial phytochrome A (CphA) and cyanobacterial phytochrome B (CphB).
