The oncoprotein MYC has the dual capacity to drive cell cycle progression or induce apoptosis, depending on the cellular context. BAG1 was previously identified as a transcriptional target of MYC that functions as a critical determinant of this cell fate decision.

Bag-1 (Bcl-2-associated athanogene) is a multifunctional anti-apoptotic protein frequently overexpressed in cancer. Bag-1 interacts with a variety of cellular targets including Hsp70/Hsc70 chaperones, Bcl-2, nuclear hormone receptors, Akt and Raf kinases.

Tetratricopeptide repeats (TPRs) are protein domains that mediate key protein–protein interactions in cells. Several TPR domains bind the C-ter-mini of the chaperones heat shock protein (Hsp)90 and⁄or Hsp70, and exchange of such binding partners is key for the heat shock response.

Although heat shock protein 70 (Hsp70) has been shown to markedly inhi-bit H2O2-induced apoptosis in C2C12 cells, and nucleolin⁄C23 has also been implicated in apoptosis, the relationship of these two molecules is still largely unknown. The aim of the current study was to investigate the potential involvement of nucleolin⁄C23 in the antiapoptotic mechanism of Hsp70.
