After the first edition published in 2007 that became a best seller, the continuous scientific developments in the field have prompted us to produce the second edition of this book. As it happens in life and science, some of the novel and promising data presented in the first edition have been confirmed, some not, and new breakthrough achievements have been accomplished. Stem Cells in Reproductive Medicine, Basic Science and Therapeutic Potential, second edition, updates the revolutionary advances in stem cell science that may potentially impact on human reproductive medicine.

Stem cell research has the potential to affect the lives of millions of people in the United States and around the world. This research is now regularly front-page news because of the controversy surrounding the derivation of stem cells from human embryos. Realizing the promise of stem cells for yielding new medical therapies will require us to grapple with more than just scientific uncertainties. The stem cell debate has led scientists and nonscientists alike to contemplate profound issues, such as who we are and what makes us human beings.

We are pleased to offer our committee’s report on guidelines for human embryonic stem cell research. This report and its recommendations are the result of many hours of committee meetings as well as a public workshop. During those sessions we heard from many dedicated and talented people who represent a wide range of views. We have tried to take these diverse perspectives into account in a report that mirrors the seriousness with which we have reflected upon them.

Twenty years after the discovery of neural stem cells, the question whether the central nervous system can be considered among regenerative tissues is still open. On one side, deep characterization of neural stem and progenitor cells, their niches, and their progeny in brain neurogenic sites overtly showed that new neurons can be generated in the brain of adult mammals, including humans.

Current procedures used for skin injury reepithelization have yet to improve. At present, stem cell-based therapies provide great potential in repairing different damaged tissues due to the multipotent character of these cells.

The ability of Ngoclinh ginseng (NLG) stem cell extract in controlling the skin aging process in human fibroblasts was investigated. To evaluate the cosmeceutical effects of the extracts on the aging process, MTT assays were conducted with various concentrations of NLG stem cells. No extracts showed any cytotoxic activities on fibroblasts at concentrations of up to 10 mg/ml.

In this study, three hASC samples from three Vietnamese people were collected and proliferated in MSCCult medium (Regemedlab, Ho Chi Minh City, Viet Nam) to the 5th cell passage. Next, hASCs were evaluated for change of mesenchymal stem cells (MSCs) characteristics including shape, immunophenotype (CD14, CD34, CD44, CD73, CD90, and/or CD166), and trilineage differentiation ability. Finally, the number of chromosomes after passages 1, 3, and 5 was evaluated by karyotyping technique.

In the present study we demonstrated that composite PPHMDS/DND coatings with elastic moduli close to those of mature bone tissue (0.2–2.8 GPa) stimulated growth and osteogenic differentiation of human adipose-derived mesenchymal stem cells (hADMSCs).

In the present study, differentiation potential of human adipose-derived stem cells (ASCs) to cardiomyocytes has been investigated by growing them on hyaluronic acid/gelatin (HA/G) plasma gels and coverslips and supplementing the growth medium with chemical modifiers (activin-a, BMP-4, insulin, valproic acid, and 5-azacytidine) in various combinations.

Electroporation is a safe and efficient method to transfect human tooth germ stem cells (hTGSCs), which are mainly mesenchymal stem cells (MSCs). In this study, pluronic F68 (PF68), a nonionic and low-foaming surfactant, was used for the first time to increase the transfection efficiency in electroporation of hTGSCs.

Human embryonic stem cells (hESCs) and reprogrammed human induced pluripotent stem cells (hiPSCs) proliferate indefinitely in the undifferentiated state (self-renewal) and differentiate into the three germ layers (and ultimately into all cell types (pluripotency).

Human endometrial stem cells (hESCs) are mesenchymal stem cells, which are responsible for the monthly renewal of the basal layer of the human endometrium by facilitating stromal and vascular regeneration. In this study, hESCs were isolated by using three different isolation methods including nonenzymatic and enzymatic digestion with trypsin and collagenase type 1.

Mesenchymal stem cells (MSCs) can be found in many types of adult tissues such as the bone marrow, adipose, placenta, liver, and periosteum. Recently, adipose tissue-derived MSCs (ADMSCs) have become one the most preferred MSC types because of their fast proliferation rate, abundance, and high plasticity for variable cell types.

Allogeneic stem cell transplantation(SCT)from a human leukocyte antigen(HLA)- matched sibling donor(MSDSCT)is the preferred first-line treatment option for young patients with severe aplastic anemia(SAA). However, only 25% of patients may find an HLA-MSD. SAA patients, who lack a suitable MSD and fail first-line immunosuppressive therapy, may consider SCT from an unrelated donor(URD-SCT)as a treatment option.

Primary graft failure is a lethal complication that occurs after allogeneic stem cell transplantation(allo-SCT)and requires retransplantation. We retrospectively assessed 1,355 patients who underwent allo-SCT at our institute. Following allo-SCT, the cumulative incidence of subsequent neutrophil engraftment was calculated each day after day 5 among patients with white blood cell(WBC)count<100 cells/μL on the respective day.
