A database containing chemical structures of 60 inhibitors and their Ki values was put into molecular operating environment (MOE) 2008.10 software, and the two-dimensional (2D) physicochemical descriptors were numerically calculated. After removing the irrelevant descriptors, a QSAR modeling was developed from the 2D-descriptors and Ki values by using the partial least squares (PLS) regression method.

The objectives of the thesis: Study on materials fabrication technology; structural - photocatalytic properties of TiO2 material, and TiO2 as the nanostructured variant. Cys341 of carboxypeptidase Y, which constitutes one side of the solvent-accessible surface of the S1 binding pocket, was replaced with Gly, Ser, Asp, Val, Phe or His by site-directed mutagenesis. Kinetic analysis, using Cbz-dipeptide substrates, revealed that polar amino acids at the 341 position increased Km whereas hydrophobic amino acids in this position tended to decrease Km.

The structure of the N-terminal docking and dimerization domain of the type IIa regulatory subunit (RIIa D/D) of protein kinase A (PKA) forms a noncovalent stand-alone X-type four-helix bundle structural motif, consisting of two helix-loop-helix monomers. RIIa D/D possesses a strong hydrophobic core and two distinct, exposed faces. A hydrophobic face with a groove is the site of protein–protein interactions necessary for subcellular localization.

Site-directedmutagenesis was used to investigate the control of 2-oxoacid cosubstrate selectivity by deacetoxycephalo-sporin C synthase. The wild-type enzyme has a requirement for 2-oxoglutarate and cannot efficiently use hydrophobic 2-oxoacids (e.g. 2-oxohexanoic acid, 2-oxo-4-methyl-penta-noic acid) as the cosubstrate. The followingmutant enzymes were produced: R258A, R258L, R258F, R258H and R258K. All of the mutants have broadened cosubstrate selectivity and were able to utilize hydrophobic 2-oxoacids.

In the yeast Saccharomyces cerevisiae, plasma membrane H + -ATPase is activated by d-glucose. We found that in the absence of glucose, this enzyme forms a complex with acetylated tubulin. Acetylated tubulin usu-ally displays hydrophilic properties, but behaves as a hydrophobic com-pound when complexed with H + -ATPase, and therefore partitions into a detergent phase.

Cyclotides are a family of bioactive plant peptides that are characterized by a circular protein backbone and three conserved tightly packed disulfide bonds. Surfactant-like membranes containing the 21-residue peptide KLLLL-KLLLLKLLLLKLLLLK (KL4 ), have been clinically tested as a therapeu-tic agent for respiratory distress syndrome in premature infants. The aims of this study were to investigate the interactions between the KL4 peptide and lipid bilayers, and the role of both the lipid composition and KL4 structure on the surface adsorption activity of KL4 -containing membranes.

K-Ras is a small G-protein, localized mainly at the inner leaflet of the plasma membrane. The membrane targeting signal of this protein consists of a polybasic C-terminal sequence of six contiguous lysines and a farnesyl-ated cysteine. Results from biophysical studies in model systems suggest that hydrophobic and electrostatic interactions are responsible for the membrane binding properties of K-Ras.

d-Crystallin is the major structural protein in avian and reptilian eye lenses, and confers special refractive properties. The protein is a homotetramer arranged as a dimer of dimers. In the present study, the roles of the side chains of Glu267, Lys315, and Glu327, which provide hydrogen bonds at the double dimer interface, were investigated. Hydrophobic side chain sub-stitution led to all mutant proteins having an unstable dimer interface.
