This study has been made in an attempt to find the structural integrity of the Chashma Nuclear power plant-1 FA skeleton at room temperature. The finite element (FE) analysis has been performed using ANSYS, in orderto determinethe elongationofthe FAskeleton as wellas the locationofmax. stress and stresses developedin axial direction under tensile load of 9800 N or 2 g being the FA handling or lifting load [Y. Zhang et al., Fuel Assembly Design Report, SNERDI, China, 1994].

(BQ) This paper reports an intelligent approach for process modeling and optimization of electric discharge machining (EDM). Physics based process modeling using finite element method (FEM) has been integrated with the soft computing techniques like artificial neural networks (ANN) and genetic algorithm (GA) to improve prediction accuracy of the model with less dependency on the experimental data.

The human b-globin gene locus is the subject of intense study, and over the past two decades a wealth of information has accumulated on how tissue-specific and stage-specific expression of its genes is achieved. The data are extensive and it would be difficult, if not impossible, to formulate a comprehensive model integrating every aspect of what is currently known. In this review, we introduce the fundamental characteristics of globin locus regulation as well as questions on which much of the current research is predicated.

The expression of mitochondrial and hydrogenosomal ADP/ATP carriers (AACs) from plants, rat and the anaerobic chytridiomycete fungus Neocallimastix spec. L2 in Escherichia coli allows a functional integration of the recombinant proteins into the bacterial cytoplasmic membrane. For AAC1 and AAC2 from rat, apparent Km values of about 40 lM for ADP, and 105 lM or 140 lM, respectively, for ATP have been determined, similar to the data reported for isolated rat mitochondria.

During the last decade several novel yeast genes encoding proteins related to the PPP family of Ser/Thr protein phosphatases have been discovered and their functional characterization initiated. Most of these novel phosphatases display intriguing structural features and/or are involved in a number of important functions, such as cell cycle regulation, protein synthesis and maintenance of cellular integrity. While in some cases these genes appear to be restricted to fungi, in others similar proteins can be found in higher eukaryotes. ...

The thermal stabilityofperoxidase fromleavesof theAfrican oil palm treeElaeis guineensis(AOPTP) at pH 3.0 was studied by differential scanning calorimetry (DSC), intrinsic fluorescence, CD and enzymatic assays. The spectral parameters asmonitoredby ellipticity changes in the far-UV CD spectrum of the enzyme as well as the increase in tryp-tophan intensity emission upon heating, together with changes in enzymatic activity with temperature were seen to be good complements to the highly sensitive but integral methodofDSC....

Leaf explants of Echinacea purpurea L. taken from aseptically germinated seedlings were inoculated with A. tumefaciens strains EHA105, carrying a binary vector conferring herbicide resistant bar gene and fungal resistant chitinase gene. Glufosinate ammonium‐resistant shoots were regenerated on a medium containing BAP and NAA at a concentration of 4.88 and 0.053 μM, respectively. A subsequent transfer of shoots to medium containing BAP was necessary for stem elongation and leaf development.

Traditional analyses of the control and regulation of steady-state concentrations and fluxes assume the activities of the enzymes to be constant. In living cells, a hierar-chical control structure connects metabolic pathways to signal-transduction and gene-expression. Consequently, enzyme activities are not generally constant. This would seem to compromise analyses of control and regulation at the metabolic level.

The topology of central carbon metabolism ofAspergillus nigerwas identified and the metabolic network reconstruc-ted, by integrating genomic, biochemical and physiological information available for this microorganism and other related fungi. The reconstructed network may serve as a valuabledatabase for annotationof genes identified in future genome sequencing projects on aspergilli. Based on the metabolic reconstruction, a stoichiometric model was set up that includes 284metabolites and335reactions,...

a2 -Macroglobulin (a2M) is an abundant glycoprotein with the intrinsic capacity for capturing diverse proteins for rapid delivery into cells. After internalization by the receptor-mediated endocytosis,a2M-protein complexes were rapidly degraded in the endolysosome system. Although this is an importantpathway for clearanceofbotha2Mandbiological targets, little is known about the nature ofa2M degradation in the endolysosome system.

BAP31 is a 28-kDa integral membrane protein of the endoplasmic reticulumwhose cytosolicdomaincontains two caspase recognition sites that are preferentially cleaved by initiator caspases, such as caspase-8. Recently,we reported that the caspase-resistant BAP31 inhibited Fas-mediated apoptotic membrane fragmentation and the release of cytochromec from mitochondria in KB epithelial cells (Nguyen M.,Breckenridge G.,Ducret A &Shore G. (2000) Mol. Cell.Biol.20,6731–6740).

ADP-ribosylation has been coupled to intracellular events associated with smooth muscle cell vasoreactivity, cytoskel-etal integrity and free radical damage. Additionally, there is evidence that ADP-ribosylation is required for smooth muscle cell proliferation. Our investigation employed selective inhibitors to establish thatmono-ADP-ribosylation and not poly(ADP-ribosyl)ation was necessary for the sti-mulationofDNAsynthesis bymitogens.

Erythrocytic stages of the malaria parasitePlasmodium fal-ciparumrely on glycolysis for their energy supply and it is unclear whether they obtain energy via mitochondrial res-piration albeit enzymes of the tricarboxylic acid (TCA) cycle appear to be expressed in these parasite stages. Isocitrate dehydrogenase (ICDH) is either an integral part of the mitochondrial TCA cycle or is involved in providing NADPH for reductive reactions in the cell.

Many pharmacologically important agents are assembled on multimodular nonribosomal peptide synthetases (NRPSs) whose modules comprise a set of core domains with all essential catalytic functions necessary for the incorporation and modification of one building block. Very often, d-amino acids are found in such products which, with few exceptions, are generated by the action of NRPS integrated epimerization (E) domains that alter the stereochemistry of the corresponding peptidyl carrier protein (PCP) bound l-intermediate....

Integrase (IN) is the retroviral enzyme responsible for the integration of the DNA copy of the retroviral genome into the host cellDNA. TheC-terminal domainof INis involved in DNA binding and enzyme multimerization. We previ-ously performed single amino acid substitutions in the C-terminal domain of the avian leukemia and sarcoma vir-uses (ALSV) IN [Moreauet al. (2002). Arch. Virol.147, 1761–1778].

One of the hallmarks of cAMP is its ability to inhibit proliferation in many cell types, but stimulate proliferation in others. Clearly cAMP has cell type specific effects and the outcome on proliferation is largely attributed to crosstalk from cAMP to the RAS⁄RAF⁄mitogen-activated protein kinase (MAPK) and extracellular signal-regulated kinase (ERK) kinase (MEK)/ ERK pathway. We review the crosstalk between these two ancient and con-served pathways, describing the molecular mechanisms underlying the interactions between these pathways and discussing their possible biological importance....

High-resolution X-ray diffraction structures of integral membrane proteins have revealed various binding modes of lipids, but current spectroscopic studies still use uniform macroscopic binding constants to describe lipid binding. The Adair approach employing microscopic lipid-binding con-stants has previously been taken to explain the enhancement of agonist binding to the nicotinic acetylcholine receptor by general anaesthetics in terms of the competitive displacement of essential lipid activator molecules [Walcher S, Altschuh J & Sandermann H (2001) J. Biol. Chem. 276, 42191–42195]....
