  • An efficient regeneration protocol was established for a rare plant species, Begonia coptidifolia. Using leaf explants, adventitious shoots were induced on Murashige and Skoog (MS) basal medium using various concentrations (1.0–10.0 mM) of three cytokinins, 6-benzyladenine (BA), thidiazuron (TDZ) and kinetin (KIN). However, adventitious shoots could not be induced on MS medium supplemented with auxins [α-naphthaleneacetic acid (NAA) or 2,4-dichlorophenoxyacetic acid (2,4-D)] tested in the 1.0–10.0 mM range.

  • The present study demonstrates a simple and efficient method for callus induction of Bamboo. The leaf, node and seed are used as explants for induction of callus. These explants were subjected to sterilization treatment and it is found that Tween -20, bavistin and ethanol are sufficient for proper sterilization of explants. Then these explants were subjected to various concentrations of growth hormone 2,4-Dichlorophenoxyacetic acid (2,4-D) added in Murashige and Skoog Media. It was concluded that the seeds are suitable explants for rapid induction of callus.

  • This paper focuses on researching of Spilanthes acmella (L.) Murr. callus culture. Different parts of the plant such as leaf, stem, flower and root have been cultured. Based on antimicrobial activity (5 - 20 mm on Staphylococcus and Vibrio) and radish root tip inhibition (82%), the callus derived from flowers is the best.

  • The present investigation was carried out in Biotechnology Laboratory, Department of Olericulture, KAU, Vellanikkara with four genotypes of cucumber comprising gynoecious, monoecious and two parthenocarpic genotypes for in vitro plant regeneration. Seeds were grown in the culture bottles quarterly filled with half strength MS basal medium. Cotyledonary leaf explants were then transferred to eight media compositions of half strength MS medium with varying IAA and BAP concentrations in the culture test tubes.

  • The aim of this study was to develop an efficient protocol to standardize the concentration of sterilants for seed and explants sterilization for in vitro germination and regeneration respectively. The explants used are cotyledonary leaf, stem and hypocotyl of in vitro grown brinjal var.KKM1. For sterilization of seeds and explants, 5% sodium hypochlorite treatment for 3 minutes is the most effective sterilant concentration against the microbiological contamination. In subsequence, the regeneration protocol was optimised for the brinjal var.KKM1.

  • In addition, the effects of explant age and lighting condition on shoot regeneration were also investigated. After 8 weeks of culture, the results showed that shoot regeneration rate (100%) and shoot multiplication coefficient (13.33) of the in vitro leaf-tTCL-4 were higher than those of other treatments and control. The shoot regeneration rate of P. edulis Sims. also varied with the change of explant age. The highest shoot regeneration rate (100%) was obtained from leaf explants of 1.5-month-old shoots after 8 weeks of culture.

  • Unexpanded leaf rolls were used as explants for callus induction with different concentrations of mutagenic chemicals (sodium nitrite and ethylmethane sulfonate). The effect of these mutagenic chemicals on callusing, shooting and rooting was studied. Callus induction was observed in different concentrations of sodium nitrite (3 mg l-1 , 5 mg l-1 and 7 mg l-1 ) and EMS (0.6 µM, 0.8 µM, 1.0 µM) along with controls. Among all the treatments 3 mg l-1 (T4) SN and 0.6 µM EMS (T10) in 2009T5 were found to be the best for callus induction traits.
