They activate p70 ribosomal protein S6 kinase (p70S6K) and acetyl-CoA carboxylase (ACC) involved in protein and fatty acids synthesis, respectively. In contrast, the AMP-activated protein kinase (AMPK), which senses the energy state of the cell and becomes activated under metabolic stress, inactivates by phosphorylation key enzymes in biosynthetic pathways thereby conserving ATP.

LTB4 12-HD catalyzes the conversion of LTB4 into 12-keto-LTB4 in the presence of NADP1, and plays an important role in inactivating LTB4.

Wilson disease (WD), an inherited disorder affecting copper metabolism, is characterized by hepatic cirrhosis and neuronal degeneration, which result from toxic levels of copper that accumulate in the liver and brain, respectively. We reported previously that the 1.3-kb promoter of the WD gene contains four metal response elements (MREs). Among the four MREs, MREa plays the most important role in the transcriptional activation of the WD promoter.

A Drosophila melanogaster cDNA clone (GH01916) encoding a putative 723-residue long (82 kDa) protein (CG 7415) and displaying 50% identity with mammalian cytosolic dipeptidyl aminopeptidase (DPP) III was func-tionally expressed in Schneider S2 cells. Immunocytochemi-cal studies using anti-(rat liver DPP III) Ig indicated the expression of this putative DPP III at the outer cell mem-brane and into the cytosol of transfected cells.

Induction of CYP3A genes by the ligand-activated preg-nane-X-receptor (PXR)involves the interaction of other as yet unidentified liver transcription factors. Here we show that the CYP3A1promoter contains two active sites con-trolled by the CCAAT/enhancer-binding protein a (C/ EBPa), previously shown to regulate a number of liver stress response genes. We have identified two functional C/EBP binding sites at theCYP3A1promoter that confer luciferase activity to C/EBPacotransfected CHO cells.

In order to study the role of peroxisome proliferator-acti-vated receptor ain mouse intestine, its agonist-induced proteins were identified by peptide mass fingerprinting fol-lowed byNorthern blot analysis using their cDNAs. One of the most remarkably induced proteins was identified as 17b-hydroxysterol dehydrogenase type 11. Its very rapid induction by various agonists was most efficient in intestine and then in liver.

Fatty acid-binding protein type 1 (FABP1), commonly termed liver-type fatty acid-binding protein (L-FABP), is encoded by a single gene in mam-mals. We cloned and sequenced cDNAs for two distinct FABP1s in zebra-fish coded by genes designatedfabp1a andfabp1b. The zebrafish proteins, FABP1a and FABP1b, show highest sequence identity and similarity to the human protein FABP1.
