This work demonstrated the potential of US for mitigating membrane fouling. However, the integration of the US transducers within the membrane system will be a major challenge for the application of US in membrane cleaning. It is more appropriate that US is used as pre-treatment to the membrane system, the transducers can be placed prior to the coagulant feed line. Further work on the determination of the detailed mechanisms associated with enhancing coagulation would enable optimisation of the system with regard to process, cost and energy efficiency.

Balçova-Narlıdere Geothermal Field (BNGF) hosts the largest geothermal district heating system of Turkey and several geothermal wells used for district heating and thermal tourism activities. This study assesses the use of BNGF geothermal fluid for agricultural activities. The spent geothermal brine was treated using nanofiltration and reverse osmosis membranes on a pilot-scale membrane test system. The qualities of the product were evaluated in terms of agricultural irrigation integrated with the implemented innovative wireless sensor network.

The aim of our in vitro study was to assess the potential effect of BPF on H295R cells mitochondrial activity, metabolic activity, membrane integrity, lysosomal function, and testosterone synthesis. Adrenocortical carcinoma cells were cultivated during 24 h in the presence of BPF (0.1, 0.5, 1, 10, 25, 50, 75, 100, 300, 500 μM). Exposure doses of BPF caused a significant decrease of mitochondrial activity starting from 1 μM, we observed a slight increase in mitochondrial activity at the lowest concentration (0.1 μM).

The expression of mitochondrial and hydrogenosomal ADP/ATP carriers (AACs) from plants, rat and the anaerobic chytridiomycete fungus Neocallimastix spec. L2 in Escherichia coli allows a functional integration of the recombinant proteins into the bacterial cytoplasmic membrane. For AAC1 and AAC2 from rat, apparent Km values of about 40 lM for ADP, and 105 lM or 140 lM, respectively, for ATP have been determined, similar to the data reported for isolated rat mitochondria.

BAP31 is a 28-kDa integral membrane protein of the endoplasmic reticulumwhose cytosolicdomaincontains two caspase recognition sites that are preferentially cleaved by initiator caspases, such as caspase-8. Recently,we reported that the caspase-resistant BAP31 inhibited Fas-mediated apoptotic membrane fragmentation and the release of cytochromec from mitochondria in KB epithelial cells (Nguyen M.,Breckenridge G.,Ducret A &Shore G. (2000) Mol. Cell.Biol.20,6731–6740).

High-resolution X-ray diffraction structures of integral membrane proteins have revealed various binding modes of lipids, but current spectroscopic studies still use uniform macroscopic binding constants to describe lipid binding. The Adair approach employing microscopic lipid-binding con-stants has previously been taken to explain the enhancement of agonist binding to the nicotinic acetylcholine receptor by general anaesthetics in terms of the competitive displacement of essential lipid activator molecules [Walcher S, Altschuh J & Sandermann H (2001) J. Biol. Chem. 276, 42191–42195]....

The ESSS protein is a recently identified subunit of mam-malian mitochondrial complex I. It is a relatively small integral membrane protein (122 amino acids) found in the b-subcomplex. Genomic sequence database searches reveal its localization to theX-chromosome inhumans andmouse. TheESSScDNAfromChinesehamster cellswas clonedand shown to complement one complementation group of our previously described mutants with a proposed X-linkage. Sequence analyses of the ESSS cDNA in these mutants revealed chain termination mutations. ...

Transport of presecretory proteins into the mammalian rough endoplasmic reticulum involves a protein translocase that comprises the integral membrane proteins Sec61ap, Sec61bp, and Sec61cp as core components. Electron microscopic analysis of protein translocase in rough micro-somal membranes suggested that between three and four heterotrimeric Sec61p complexes form the central unit of protein translocase.

TheCa 2+ -ATPaseof skeletalmuscle sarcoplasmic reticulum (SERCA), an integral membrane protein, becomes irre-versibly inactivatedin vitroby the addition of a single bolus of peroxynitritewithaK0.5of 200–300lM, and this results in a large decrease of theATP-dependentCa 2+ gradient across the sarcoplasmic reticulum (SR) membranes. The inactiva-tion of SERCA is raised by treatment of SR vesicles with repetitivemicromolar pulses of peroxynitrite.

Báo cáo khóa học: High level cell-free expression and specific labeling of integral membrane proteins
We demonstrate the high level expression of integral membrane proteins (IMPs) in a cell-free coupled tran-scription/translation system using a modified Escherichia coli S30 extract preparation and an optimized protocol. The expression of theE. colismall multidrug transporters EmrE and SugE containing four transmembrane segments (TMS), the multidrug transporter TehA with 10 putative TMS, and the cysteine transporter YfiK with six puta-tive TMS, were analysed.

Cell-free expression has become a highly promising tool for the fast and efficient production of integral membrane proteins. The proteins can be produced as precipitates that solubilize in mild detergents usually without any prior denaturation steps. Alternatively, membrane proteins can be syn-thesized in a soluble form by adding detergents to the cell-free system.

Endocytic and biosynthetic trafficking pathways to the lysosome⁄vacuole converge at the prevacuolar endosomal compartment. During transport through this compartment, integral membrane proteins that are destined for delivery to the lysosome⁄vacuole lumen undergo multivesicular body (MVB) sorting into internal vesicles formed by invagination of the endo-somal limiting membrane.

Mitochondria evolved from a bacterial endosymbiont ancestor in which the integral outer membrane proteins would have been b-barrel structured within the plane of the membrane. Initial proteomics on the outer mem-brane from yeast mitochondria suggest that while most of the protein components are integral in the membrane, most of these mitochondrial pro-teins behave as if they have a-helical transmembrane domains, rather than b-barrels.

Alr1p is an integral plasma membrane protein essential for uptake of Mg2+ into yeast cells. Homologs of Alr1p are restricted to fungi and some protozoa. Alr1-type proteins are distant relatives of the mitochondrial and bacterial Mg 2+ -transport proteins, Mrs2p and CorA, respectively, with which they have two adjacent TM domains and a short Mg 2+ signature motif in common.

The mitochondrial respiratory complex II, or succinate:ubiquinone oxido-reductase, is an integral membrane protein complex in both the tricarboxy-lic acid cycle (Krebs cycle) and aerobic respiration. The gene sequences of each complex II subunit were measured by RT-PCR. N-terminal sequen-cing work was performed to identify the mitochondrial targeting signal peptide of each subunit.
