A comparative study of thermodynamic and kinetic aspects of Cu(II) and Ni(II) binding at the N-terminal binding site of human and bovine serum albumins (HSA and BSA, respectively) and short peptide analogues was performed using potentiometry and spectroscopic techniques. It was found that while qualitative aspects of interaction (spectra and structures of complexes, order of reactions) could be reproduced, the quantitative parameters (stability and rate constants) could not.

The highly conserved, atypical RIO serine protein kinases are found in all organisms, from archaea to man. In yeast, the kinase activity of Rio2 is necessary for the final processing step of maturing the 18S ribosomal rRNA. We have previously shown that the Rio2 protein from Archaeo-globus fulgidus contains both a small kinase domain and an N-terminal winged helix domain.

We studied the interaction of several metal ions with the copper chaperone fromEnterococcus hirae(EhCopZ). We show that the stoichiometry of the protein–metal complex varies with the experimental conditions used. At high con-centration of the protein in a noncoordinatingbuffer, a dimer, (EhCopZ)2–metal, was formed. The presence of a potentially coordinatingmolecule L in the solution leads to the formation of a monomeric ternary complex, EhCopZ– Cu–L, where L can be a buffer or a coordinatingmole-cule (glutathione, tris(2-carboxyethyl)phosphine). ...
