Molecular characterization of fusarium

Potato dry rot disease (Fusarium spp.) is one of the most important fungal storage rots on potato tubers after harvesting. The aims of this study were to identify the Fusarium species that cause dry rot in potato storages, evaluate the most virulent Fusarium species in vitro and in vivo for fungicide sensitivity and determine the susceptibility of the cultivars. In this context, as a result of surveys carried out in potato storages of Ödemiş and Torbalı counties in İzmir province in 2015–2016; Fusarium species obtained from diseased potato tubers were identified as Fusarium sambucinum, F.

Pokkah boeng is one of the most serious and devastating diseases of sugarcane and causes significant loss in cane yield and sugar content. Although carbendazim is widely used to prevent fungal diseases, the molecular basis of Fusarium species complex (FSC) resistance to carbendazim remains unknown.

Maize ear rot caused by F. verticillioides which is prevalent on maize under Punjab conditions. The symptoms appear as white-pink fungal growth and causes “starburst” pattern of white streaks on the kernels. Thirty Fusarium isolates were isolated from the infected maize plant from different agro-chemical zones of Punjab.

Stalk rot complex of maize (Zea Mays L.) caused by Macrophomina phaseolina (Tassi) Goid and Fusarium verticilloides (Sacc) Nirenberg reduces yield directly by affecting the physiological activity of the plants and finally cause lodging, which is the main reason of economic losses. Screening of Indian maize inbreds for the resistance to pathogen was done using 34 Simple Sequence Repeats (SSR) markers available in database.

The study was undertaken to isolate the DNA of good quality from the banana wilt pathogen Fusarium oxysporum f. sp. cubense (Foc) directly from infected tissues for molecular studies. In this method, laborious and time-consuming procedures such as isolation of the pathogen and its artificial cultivation are avoided. Moreover, grinding of the fungal mycelium using liquid nitrogen is also not needed as the DNA is directly extracting from infected banana tissues.

Bitter gourd (Momordica charantiaL.) is an important cucurbitaceous plant in which the deleterious diseases namely vascular wilt, damping off occurs commonly and causes 30 to 50 percent losses in crop. The present investigation was aimed to study the morphological and molecular characterization of disease causing pathogen to understand the etiology of the disease. Survey was conducted at different locations of Coimbatore, Erode and Dharmapuri and samples were collected for the isolation of the pathogen.

Fusarium species cause considerable crop losses worldwide and necessitate an expensive use of chemical pesticides. Resistant genes can be used to improve plant resistance through gene editing and gene silencing strategies. Currently, information on host defences against Fusarium species is limited. Through in-silico tools, identification of genes and understanding of disease resistance mechanism has become possible.

On the basis of cultural, morphological, molecular characteristics and pathogenicity test, the fungus was confirmed as F. oxysporum Schlechtend. Fr. f. sp. ciceri (Padwick) Matuo and K. Sato. The pathogenic nature of fifteen isolates tested on chickpea wilt susceptible cultivar JG-62, two isolate (Char, Choki) were found non-pathogenic gave zero per cent disease incidence (PDI), while one isolate (Chittal) found highly pathogenic with 100 per cent PDI which was further used for molecular identification and screening of agro-chemicals.

Vanilla planifolia is popularly known as Prince of Spices a fleshy perennial liana grown in and around Western Ghats for its natural compounds used in several ice creams, chocolates and beverages. Fusarium oxysporum f.sp. vanillae is one of the most destructive pathogens causing severe loss to yield and during the survey conducted in 2016, maximum of 25 % incidence was noticed in coorg district. The pathogen was isolated and morphologically identified as F. oxysporum based on the conidial, chlamydosporial and cultural characters. The size of microconidia ranged between 5.97 to 8.

Regionally grown banana cultivars are severely infected by several pathogens one such Fusarium oxysoporum f. sp. Cubense which causes significant crop losses in the fields. In this study fusarium wilt screening was carried out on gamma ray induced mutants and molecular characterization was done to assess the acquired resistance to Fusarium oxysoporum f. sp. cubense. From the screening we observed that gamma ray derived mutants are more sensitive in 25, 30 and 35 Gy. Whereas, several mutants obtained from 40 and 45 were resistant as comparable with untreated control.
