The current research project genotyped HAV RNA by using protocols and primer sets designed by HAVNET. It collected and genotyped previous HAV-positive samples in VIDRL sample bank, and the results were used to establish epidemiological and molecular database of the HAV strains. The database was used to investigate the relationship between the current diagnostic samples and the previous HAV infection cases. It was expected to identify the source of the hepatitis A infection.

A database containing chemical structures of 60 inhibitors and their Ki values was put into molecular operating environment (MOE) 2008.10 software, and the two-dimensional (2D) physicochemical descriptors were numerically calculated. After removing the irrelevant descriptors, a QSAR modeling was developed from the 2D-descriptors and Ki values by using the partial least squares (PLS) regression method.

As a first step toward developing a zebrafish model for investigating the role of sulfation in counteracting environ-mental estrogenic chemicals, we have embarked on the identification and characterization of cytosolic sulfotrans-ferases (STs) in zebrafish. By searching the zebrafish expressed sequence tag database, we have identified two cDNA clones encoding putative cytosolic STs. These two zebrafish ST cDNAs were isolated and subjected to nuc-leotide sequencing. The 2-hydroxycarboxylate transporter (2HCT) family of secondary trans-porters belongs to a much larger structural class of secondary transporters termed ST3 which contains about 2000 transporters in 32 families. The transporters of the 2HCT family are among the best studied in the class. The membrane associated proteins in eicosanoid and glutathione metabo-lism (MAPEG) superfamily includes structurally related membrane proteins with diverse functions of widespread origin. A total of 136 proteins belong-ing to the MAPEG superfamily were found in database and genome screenings. The members were found in prokaryotes and eukaryotes, but not in any archaeal organism.

A novel cytochrome P450, CYP4x1, was identified in EST databases on the basis of similarity to a conserved region in the C-helix of the CYP4A family. The human and mouse CYP4x1 cDNAs were cloned and found to encode putative cytochrome P450 proteins. Molecular modelling of CYP4x1 predicted an unusual substrate binding channel for the CYP4 fam-ily.

Four cDNA clones that were annotated in the database as encoding d-amino acid oxidase (DAAO) or d-aspartate oxidase (DASPO) were iso-lated by RT-PCR from Caenorhabditis elegans RNA. The proteins (Y69Ap, C47Ap, F18Ep, and F20Hp) encoded by the cloned cDNAs were expressed in Escherichia colias recombinant proteins with an N-terminal His-tag.
