This paper describes the cloning of the genes coding for each component of the complex of toluene/o-xylene monooxy-genase fromPseudomonas stutzeriOX1, their expression, purification and characterization. Moreover, the reconsti-tution of the active complex from the recombinant subunits has been obtained, and the functional role of each compo-nent in the electron transfer from the electron donor to molecular oxygen has been determined.

The catalytic activity of phenylalanine hydroxylase (PAH, phenylalanine 4-monooxygenase EC 1.14.16.1) is regulated by three main mechanisms, i.e. substrate (L-phenylalanine, L-Phe) activation, pterin cofactor inhibition and phos-phorylation of a single serine (Ser16) residue. To address the molecular basis for the inhibition by the natural cofactor (6R)-L-erythro-5,6,7,8-tetrahydrobiopterin, its effects on the recombinant tetrameric human enzyme (wt-hPAH) was studied using three different conformational probes, i.e.

Soluble methane monooxygenase (sMMO) is a three-com-ponent enzyme that catalyses dioxygen- and NAD(P)H-dependent oxygenation of methane and numerous other substrates. Oxygenation occurs at the binuclear iron active centre in the hydroxylase component (MMOH), to which electrons are passed from NAD(P)H via the reductase component (MMOR), along a pathway that is facilitated and controlledby the third component, proteinB (MMOB).

Alkene monooxygenase (AMO) fromRhodococcus rhodochrous(formerly Nocardia corallina) B-276 belongs to a family of multicomponent nonheme binuclear iron-centre oxygenases that includes the soluble methane mono-oxygenases (sMMOs) found in some methane-oxidizing bacteria. The enzymes catalyse the insertion of oxygen into organic substrates (mostly hydrocarbons) at the expense of O2and NAD(P)H.

Planarians are one of the simplest animal groups with a central nervous system. Their primitive central nervous system produces large quantities of a variety of neuropeptides, of which many are amidated at their C terminus. In vertebrates, peptide amidation is catalyzed by two enzymes [peptidylglycine a-hydroxylating monooxygenase (PHM) and peptidyl-a-hydroxylglycinea-amidating lyase] acting sequentially. In mammals, both enzymatic activities are contained within a single protein that is encoded by a single gene....

The Baeyer–Villiger monooxygenase (BVMO), 4-hydroxy-acetophenone monooxygenase (HAPMO), uses NADPH andO2 to oxidize a variety of aromatic ketones and sulfides. The FAD-containing enzyme has a 700-fold preference for NADPH over NADH. Sequence alignment with other BVMOs, which are all known to be selective for NADPH, revealed three conservedbasic residues, whichcouldaccount for the observed coenzyme specificity. The corresponding residues in HAPMO (Arg339, Lys439 and Arg440) were mutated and the properties of the purified mutant enzymes were studied. ...

Vitamin C, a reducing agent and antioxidant, is a cofactor in reactions cata-lyzed by Cu + -dependent monooxygenases and Fe 2+ -dependent dioxygenas-es. It is synthesized, in vertebrates having this capacity, fromd-glucuronate. The latter is formed through direct hydrolysis of uridine diphosphate (UDP)-glucuronate by enzyme(s) bound to the endoplasmic reticulum mem-brane, sharing many properties with, and most likely identical to, UDP-glucuronosyltransferases.
