This research uses empirical data from a cross-sectional survey of households that was specifically designed to examine the vulnerability and resilience of households to global macroeconomic shocks. In early 2011 six communities in each country were surveyed, ranging from inner-urban squatter settlements of the respective capital cities to some of the most geographically distant areas in each country. The original contribution of this thesis is that it marks the first time that the micro effects of macroeconomic events are studied in such a broad range of communities in either country.

The K103N substitution is a frequently observed HIV-1 RT mutation in patients who do not respond to combinationtherapy. The drugs Efavirenz, MSC194 and PNU142721 belong to the recent generation of NNRTIs characterized by an improved resistance profile to the most common single point mutations within HIV-1 RT, including the K103N mutation. In the present study we present structural observations from Efavirenz in complex with wild-type protein and the K103N mutant and PNU142721 and MSC194 in complex with the K103N mutant.

Coenzyme F420 is a 5-deazaflavin. Upon reduction, 1,5 dihydro-coenzyme F420 is formed with a prochiral centre at C5. All the coenzyme F420-dependent enzymes investigated to date have been shown to be Si-face stereospecific with respect to C5 of the deazaflavin, despite most F420 -dependent enzymes being unrelated phylogenetically.
