A series of trehalose-based oligosaccharides were isolated from the cytoplasmic fraction of Mycobacterium smegmatis and purified by gel-filtration and paper chromatography and TLC. Their structures were determined by HPLC and GLC to determine sugar composition and ratios, MALDI-TOF MS to measure molecular mass, methylation analysis to determine linkages, 1H-NMR to obtain anomeric configurations of glycosidic linkages, and exoglycosidase digestions followed by TLC to determine sequences and anomeric configurations of the monosaccharides....

Trehalose synthase (TreS) catalyzes the reversible inter-conversion of trehalose (glucosyl-a,a-1,1-glucose) and maltose (glucosyl-a1-4-glucose). TreS was purified from the cytosol ofMycobacterium smegmatisto give a single protein band on SDS gels with a molecular mass of68 kDa. However, active enzyme exhibited a molecular mass of 390 kDa by gel filtration suggesting that TreS is a hexa-mer of six identical subunits.

Trehalose is a nonreducing disaccharide of glucose (a,a-1,1-glucosyl-glu-cose) that is essential for growth and survival of mycobacteria. These organisms have three different biosynthetic pathways to produce trehalose, and mutants devoid of all three pathways require exogenous trehalose in the medium in order to grow.

CmtR is a repressor of the ArsR–SmtB family fromMycobacterium tuber-culosis that has been shown to sense Cd(II) and Pb(II) in Mycobacte-rium smegmatis. We establish here that CmtR binds cooperatively to multiple sites inM. tuberculosisDNA and protects an unusually long 90 bp AT-rich sequence from )80 bp to +10 with respect to its own initiation codon.

Two structures of monomeric methionyl-tRNA synthetase, fromMycobac-terium smegmatis, in complex with the ligands methionine⁄adenosine and methionine, were analyzed by X-ray crystallography at 2.3 A˚ and at 2.8 A˚ , respectively. The structures demonstrated the flexibility of the multidomain enzyme. A new conformation of the structure was identified in which the connective peptide domain bound more closely to the catalytic domain than described previously.
