Mycobacterium smegmatis

The essential oil (EO) of Cota tinctoria var. tinctoria was analyzed using GC-FID / MS. A total of 51 compounds were determined from this taxon, accounting for 99.79% in hydrodistillation. Monoterpenes were the primary chemical class for the volatile organic compounds in the EO (36.1%, 13 compounds). Borneol (18.1%), camphor (14.9%), and β-pinene (11.3%) were the major components in the EO of C. tinctoria var. tinctoria.

In the present work, the volatiles of mosses [Hylocomium splendens (Hedw.) Schimp. (Hylocomiaceae) and Leucodon sciuroides (Hedw.) Schw¨agr. (Leucodontaceae)] have been investigated by GC-FID and GC/MS. Fifty-eight compounds in the oil of H. splendens, representing 75.4%, and 41 compounds in the oil of L. sciuroides, representing 87.6%, were identified. The major components were found to be β -pinene (11.6%) and α-pinene (8.9%) in the oil of H. splendens, and nonanal (26.8%) and heptanal (13.7%) in the oil of L. sciuroides.

The new compounds 7a–k, 8a–k, and 9a–k were synthesized from flavonones 4–6, which can be considered new precursors for quinoline synthesis through a one-step reaction. All the target compounds (7a–k, 8a–k, and 9a–k) were evaluated for their in vitro antimicrobial activity against nine test microorganisms. They showed the most activity against Mycobacterium smegmatis with minimum inhibitory concentrations (MIC) of 62.5–500 µg/mL, indicating their potential uses as antituberculosis agents. Among them 8a–k (m-fluoride) were the most active compounds against M. smegmatis (MIC, 62.5–125 µg/mL).

Twenty-five new hydroxy- and methoxy-substituted 4,6-diarylpyrimidin-2(1H)-ol (20–34) and 4,6-diarylpyrimidine-2(1H)-thiol derivatives (35–44) were synthesized from the reaction of the corresponding 1,3-diaryl-2-propene-1- one compounds (1–19) with urea or thiourea using the solid-phase microwave method. All the new synthetic compounds (20–44) were evaluated with regard to their α-glucosidase activity. However, only compounds 22–25, 27, 31, 34, 35, 37, and 40 exhibited a greater inhibitory effect than standard acarbose. The IC50 values of the active compounds ranged between 2.36 and 13.

A series of 3-mercapto-1,2,4-triazole–pyrrole hybrids was designed as antimycobacterial agents by employing 5- (4-(1H -pyrrol-1-yl)phenyl)-4H -1,2,4-triazole-3-thiol as the scaffold onto which several types of moieties were introduced in the triazole ring at N-4 and N-2 and as substituents of the mercapto function. The aforementioned moieties are an allyl or a phenyl moiety at N-4; an aminomethyl group at N-2; or methyl, substituted benzyl, ethoxycarbonylmethyl, or substituted phenacyl at sulfur.

The volatile components of essential oil (EO), SPME, and SPME of solvent extracts (n-hexane, methanol, and water) obtained from fresh Serapias orientalis subsp. orientalis(Soo) were analyzed by GC-FID/MS. EO of Soo gave 11 compounds in the percentage of 99.97%; capronaldehyde (37.01%), 2-(E)-hexenal (23.19%), and n-nonanal (19.05%) were found to be major constituents. The methanolic extracts had antibacterial activity against Aeromonas hydrophila, Bacillus brevis, B. cereus, Klebsiella pneumoniae, Pseudomonas aeruginosa, and Staphylococcus aureus ATCC 29213. However, no activity was found against Escherichia coli, Morganella morganii, Mycobacterium smegmatis, Proteus mirabilis, Yersinia enterocolitica, or Saccharomyces cerevisiae.

A series of trehalose-based oligosaccharides were isolated from the cytoplasmic fraction of Mycobacterium smegmatis and purified by gel-filtration and paper chromatography and TLC. Trehalose synthase (TreS) catalyzes the reversible inter-conversion of trehalose (glucosyl-a,a-1,1-glucose) and maltose (glucosyl-a1-4-glucose). TreS was purified from the cytosol ofMycobacterium smegmatisto give a single protein band on SDS gels with a molecular mass of68 kDa. However, active enzyme exhibited a molecular mass of 390 kDa by gel filtration suggesting that TreS is a hexa-mer of six identical subunits.

Two structures of monomeric methionyl-tRNA synthetase, fromMycobac-terium smegmatis, in complex with the ligands methionine⁄adenosine and methionine, were analyzed by X-ray crystallography at 2.3 A˚ and at 2.8 A˚ , respectively. The structures demonstrated the flexibility of the multidomain enzyme. A new conformation of the structure was identified in which the connective peptide domain bound more closely to the catalytic domain than described previously.
