This thesis documents a research program in which Australian grown Danshen extracts and related bioactive compounds have been demonstrated to be effective in scavenging free radicals, namely ROS generation, in a cell free system and NADPH oxidase derived superoxide (O2-) generation in vascular tissue (rat aorta). Danshen extracts, both aqueous and ethanol, and their active compounds (Tanshinone IIA, Salvianolic Acid B and Danshensu) demonstrated anti-oxidant activity however findings exhibited stronger activity in a cell free system than in the aortic vascular tissue.

Among these, salinity is a worldwide problem that impacts the fertility of arable lands and sustainability of food security, which is getting more attention due to climate change. Halophytes can survive and reproduce in soils containing high concentrations of salt and have developed adaptation mechanisms at physiological, biochemical, and molecular levels including maintenance of ROS metabolism.

In a previous study, the S100A8/A9 protein, a Ca2+- and arachidonic acid-binding protein, abundant in neutrophil cytosol, was found to potentiate the activation of the redox component of the O2– generating oxidase in neutrophils, namely the membrane-bound flavocytochrome b, by the cytosolic phox proteins p67phox, p47phox and Rac ` (Doussiere J., Bouzidi F. and Vignais P.V. (2001) Biochem. Biophys. Res. Commun. 285, 1317–1320). This led us to check by immunoprecipitation and protein fractionation whether the cytosolic phox proteins could bind to S100A8/A9.
