Upstream open reading frames (uORFs) in the 5′-untranslated regions (5′-UTRs) of certain eukaryotic mRNAs encode evolutionarily conserved functional peptides, such as cis-acting regulatory peptides that control translation of downstream main ORFs (mORFs).

The cell line, RCS-LTC (derived from the Swarm rat chondrosarcoma), deposits a copious extracellular matrix in which the collagen component is primarily a polymer of partially processed type II N-procollagen molecules. Transmission electron microscopy of the matrix shows no obvious fibrils, only a mass of thin unbanded filaments. We have used this cell system to show that the type II N-pro-collagen polymer nevertheless is stabilized by pyridinoline cross-links at molecular sites (mediated by N- and C-telo-peptide domains) found in collagen II fibrils processed nor-mally....

High-resolution crystal structures of large ribosomal subunits from Deinococcus radioduranscomplexed with tRNA-mimics indicate that precise substrate positioning, mandatory for efficient protein biosynthesis with no further conformational rearrangements, is governed by remote interactions of the tRNA helical features.

Ribosomes position their substrate at stereochemistry suitable for peptide bond formation, and promote substrate-mediated cata-lysis. The linkage between substrate orientation, dominated by remote interactions, and a sizable symmetrical region identified in all known ribosome structures indicates a guided rotatory motion of aminoacylated-tRNAs along a ribosomal path leadings to the advance of nascent peptides into the protein exit tunnel at an extended conformation.

Twin-arginine translocation (Tat)-mediated protein trans-port across the bacterial cytoplasmicmembrane occurs only after synthesis and folding of the substrate protein that contains a signal peptide with a characteristic twin-arginine motif. This implies that premature contact between the Tat signal peptide and the Tat translocon in themembranemust be prevented. We used site-specific photo-crosslinking to demonstrate that the signal peptide of nascent Tat proteins is in close proximity to the chaperone and peptidyl-prolyl isomerase trigger factor (TF). ...

Mycobacterium tuberculosisis the causative agent of human tuberculosis (TB). Mycobacterial secretory protein ESAT-6 induces matrix metallopro-teinase (MMP)-9 in epithelial cells neighboring infected macrophages. MMP-9 then enhances recruitment of uninfected macrophages, which con-tribute to nascent granuloma maturation and bacterial growth.
