Xanthomas are well-circumscribed lesions in the connective tissue of the skin, tendons, or fasciae that predominantly consist of foam cells; these specific cells are formed from macrophages as a result of an excessive uptake of low-density lipoprotein (LDL) particles and their oxidative modification.

It is widely accepted that oxidative modification of lowdensity lipoprotein (LDL) plays a pivotal role in the initiation and development of atherosclerosis. In the present study, we found that the MeOH and H2O extracts of the plant Ampelopsis cantoniensis, and its main constituent, myricetin, possessed significant protective effects on LDL oxidation induced by either a metal ion (Cu2+) or a free radical (AAPH).

We reported earlier that urate may behave as a pro-oxidant in Cu 2+ -induced oxidation of diluted plasma. Thus, its effect on Cu 2+ -induced oxidation of isolated low-density lipoprotein (LDL) was investigated by mon-itoring the formation of malondialdehyde and conjugated dienes and the consumption of urate and carotenoids. We show that urate is antioxidant at high concentration but pro-oxidant at low concentration.

The beneficial influence of dietary curcumin, capsaicin and their combina-tion on the susceptibility of low-density lipoprotein (LDL) to oxidation was examined in an animal study. Individually, both dietary curcumin and capsaicin significantly inhibited thein vivo iron-induced LDL oxidation, as well as copper-induced oxidation of LDLin vitro. The protective effect of the combination of curcumin and capsaicin on LDL oxidation was greater than that of individual compounds.

We recently reported that, depending on its concentration, urate is either apro- or anantioxidant inCu 2+ -induced low-density lipoprotein (LDL)oxidation. We also previously demonstrated an antioxidant synergy between urate and some flavonoids in the Cu 2+ -induced oxidation of diluted serum. As a result, the effectof theflavonoidquercetinon the Cu 2+ -induced oxidation of isolated LDL has been studied either in the presence or absence of urate.

Neurodegenerative disorders are associated with oxidative stress. Low den-sity lipoprotein (LDL) exists in the brain and is especially sensitive to oxi-dative damage. Oxidative modification of LDL has been implicated in the pathogenesis of neurodegenerative diseases. Therefore, protecting LDL from oxidation may be essential in the brain.

Proprotein convertase subtilisin⁄kexin type 9 (PCSK9) interferes with the recycling of low-density lipoprotein (LDL) receptor (LDLR). This leads to LDLR degradation and reduced cellular uptake of plasma LDL. Naturally occurring human PCSK9 loss-of-function mutations are associated with low levels of plasma LDL cholesterol and a reduced risk of coronary heart disease.

Tribble3 (TRB3), a mammalian homolog ofDrosophilatribbles, slows cell-cycle progression, and its expression is increased in response to various stresses. The aim of this study was to investigate the role of theTRB3gene in macrophage apoptosis induced by oxidized low-density lipoprotein (ox-LDL). We found that, in human monocyte-derived macrophages, TRB3is upregulated by ox-LDL in a dose- and time-dependent manner.

Many in vitro studies have clearly revealed the potent role of flavonoids in inhibiting lipid peroxidation and oxidation of low-density lipoproteins (LDL). It has been proposed that flavonoids near membrane surfaces are ideally located for scavenging free radicals generated in the aqueous phase.
