Phospholipase A2 receptor

The M-type phospholipase A2 receptor (PLA2R1) plays a crucial role in several signaling pathways and may act as tumor-suppressor. This study examined the expression and methylation of the PLA2R1 gene in Jurkat and U937 leukemic cell lines and its methylation in patients with myelodysplastic syndrome (MDS) or acute leukemia.

It has recently been proposed that the M-type phospholipase A2 receptor (PLA2R1) acts as a tumour suppressor in certain malignancies including mammary cancer. Considering that DNA methylation is an important regulator of gene transcription during carcinogenesis, in the current study we analyzed the PLA2R1 expression, PLA2R1 promoter methylation, and selected micro RNA (miRNA) levels in normal human mammary epithelial cells (HMEC) and cancer cell lines.

Insect attack-wound in plant cells stimulate huge number of signal molecules to induce and activate defense system. Among these one of the intracellular signal molecules are small peptide systemin (18AAs long) produced by proteolytic cleavage in wounded cells from prosystemin (200As) precursor protein. Systemin then bind to its SR-160 (Systemin Cell Surface Receptor) protein (160 kD) and activate phospholipase A2 (PLA-2) that releases linolenic acid, which is initial precursor for JA biosynthesis.

According to recent studies, the phospholipase A2 receptor 1 (PLA2R1) may be used as a biomarker to diagnose idiopathic membranous nephropathy (iMN). Moreover, the immune-dominant regions of PLA2R1 have been identified.

Ammodytoxins (Atxs) are presynaptically acting snake venom phospholipase A2 (PLA2) toxins the molecular mechanism of whose neurotoxicity is not completely understood. Two chimeric PLA2s were prepared by repla-cing the C-terminal part of a nontoxic venom PLA2 , ammodytin I2, with that of AtxA(K108N). The chimeras were not toxic, but were able to bind strongly to an Atxs-specific neuronal receptor, R25.

Comparison of the crystal structures of threeMicropechis ikahekaphos-pholipase A2 isoenzymes (MiPLA2, MiPLA3 and MiPLA4, which exhibit different levels of pharmacological effects) shows that their C-terminus (residues 110–124) is the most variable. M-Type receptor binding affinity of the isoenzymes has also been investigated and MiPLA4 binds to the rabbit M-type receptor with high affinity. Examination of surface charges of the isoenzymes reveals a trend of increase in positive charges with potency....

This diverse collection of research articles is united by the enormous power of modern molecular genetics. Every author accomplished two objectives: (1) making the field and the research described accessible to a large audience and (2) explaining fully the genetic tools and approaches that were used in the research. One fact stands out - the importance of a genetic approach to addressing a problem. I encourage you to read several chapters.
