  • Seed propagation of Mycetia balansae Drake is not easy, due to low germination rate and low viability, so it is very rare in nature. In order to conserve this plant as well as to use for commercial purposes in cooperation with Tue Linh Co.Ltd, providing a large number of plants is necessary and in vitro tissue culture technology could be solve this problem. Therefore, the study on "Callus induction and plant regeneration in vitro from leaf explants of mycetia balansae drake" was carried out.

  • The in vitro raised plantlets were successfully acclimatized under ex vitro conditions and grew well in the glasshouse. All the regenerated plantlets appeared normal with respect to morphological characteristics with 100% survival rate. Somaclonal variation in tissue culture is a common phenomenon which makes it mandatory to check for genetic stability of plants. The genetic variation in the in vitro raised plants was confirmed using Inter Simple Sequence Repeats(ISSR) markers. Comparison of the bands with the mother plant revealed that the genetic variation frequency reached 6.

  • Paphiopedilum orchids, the most popular and rare orchid genera, are used as potted flowers and cut flowers. In nature condition, the orchid population is threatened with extinction due to over-exploitation and altered habitat. Reduction in this orchids via large-scale micropropagation is a better option to reduce the wild harvest, meets the commercial needs as well as conserves this threatened orchid. This orchid can be propagated by germinating seeds under in vitro culture condition; however, they are difficult for micropropagation, especially regenerating plantlets from in vitro culture.

  • In vitro regeneration and in planta transformation protocols were standardized for the black gram (Vigna mungo L.) cv. VBN 3, an important grain legume. An efficient regeneration protocol was developed using 1-week-old de-embryonated cotyledon as explants. The maximum frequency of shoot regeneration (72.6%), multiple shoot induction (6.0 ± 0.3) and shoot elongation was achieved on MS medium supplemented with 3.0 mg/l 6-benzyl amino purine (BAP) after 4 weeks of culture. Up to 65% of the regenerated shoots were rooted on MS medium containing 0.

  • The present study was carried out to monitor somaclonal variations of banana cv. Ney Poovan (AB) during micropropagation using random amplified polymorphic DNA (RAPD). Sword suckers were used as explants and regenerated on MS media supplemented with BAP alone, BAP + Kinetin and TDZ at varying levels of concentrations. The regenerated shoots were again subcultured on MS medium fortified with plant growth regulators at different concentrations. Rooting was obtained on ½ MS medium supplemented with 2.00 mg/l IBA and 2 g/l of activated charcoal.

  • Micropropagation is a highly sought after technique in the commercial production of orchid plants. This has the advantage of providing large number of plants in a short period of time. But a major constrain in the in vitro propagation technique is the somaclonal variation. It is important to produce true to type planting materials especially in case of the hybrids as they are more prone to variations. The true to type nature of the micropropagated plantlets can be confirmed by genetic fidelity analysis.

  • An in vitro protocol for plant regeneration through organogenesis was established for Picrorhiza kurroa Royle Ex. Benth (Scrophulairaceae). The plant is a principle source of glycoside that is Picroside-I, Picroside-II and Kutkoside. Callus was induced on Murashige and Skoog basal medium supplemented with cytokinin (Kn or BA) and auxin (2,4-D/IBA/NAA) using leaf, petiole and stem explants. Shoots were regenerated on MS media with BA (1.5 to 2.5 mgl-1 ) combined with auxin (IBA or NAA) in the range of 0.5 to 2.5 mgl-1 . Rooting was initiated with full or half media supplemented with 0.

  • Coconut has a wide range of applications from culinary to industrial areas. Loss of coconut trees due to the devastating effect of deadly diseases and the natural aging of the plants demand rejuvenation of orchards with elite seedlings. A conventional breeding program is not sufficient to meet the growing need for healthy planting materials. However, in vitro culture studies can pave the way for mass multiplication. In this study, the effect of TDZ on sliced mature embryo explants through direct organogenesis of coconut var. Chowghat Orange Dwarf (COD) was investigated.
