Seed propagation of Mycetia balansae Drake is not easy, due to low germination rate and low viability, so it is very rare in nature. In order to conserve this plant as well as to use for commercial purposes in cooperation with Tue Linh Co.Ltd, providing a large number of plants is necessary and in vitro tissue culture technology could be solve this problem. Therefore, the study on "Callus induction and plant regeneration in vitro from leaf explants of mycetia balansae drake" was carried out.

Tissue culture and rapid propagation technology is an important way to solve the difficulties of plant propagation. This experiment aims to explore the appropriate conditions at each stage of the red maple’s tissue culture process and to obtain plantlets, thus providing a theoretical basis for the establishment of the red maple’s tissue culture system.

The in vitro raised plantlets were successfully acclimatized under ex vitro conditions and grew well in the glasshouse. All the regenerated plantlets appeared normal with respect to morphological characteristics with 100% survival rate. Somaclonal variation in tissue culture is a common phenomenon which makes it mandatory to check for genetic stability of plants. The genetic variation in the in vitro raised plants was confirmed using Inter Simple Sequence Repeats(ISSR) markers. Comparison of the bands with the mother plant revealed that the genetic variation frequency reached 6.

Studies were conducted to know varietal response on plantlet regeneration through macropropagation in banana. Many varieties and cultivars were grown in different regions of country and there is a need to know which variety will respond well to macro-propagation. Three commercially cultivated varieties were taken for experiment viz., Ney Poovan, Rajapuri and Grand Naine.

Paphiopedilum orchids, the most popular and rare orchid genera, are used as potted flowers and cut flowers. In nature condition, the orchid population is threatened with extinction due to over-exploitation and altered habitat. Reduction in this orchids via large-scale micropropagation is a better option to reduce the wild harvest, meets the commercial needs as well as conserves this threatened orchid. This orchid can be propagated by germinating seeds under in vitro culture condition; however, they are difficult for micropropagation, especially regenerating plantlets from in vitro culture.

Liquid Murashige and Skoog (MS) medium in varying strengths, is the most commonly utilized medium for proficient rooting in the regenerated plants in doubled haploidy breeding protocols. On the other side, CPV (cocopeat: perlite: vermiculite) medium is currently being used in hardening of tissue culture regenerated plants.

In vitro regeneration and in planta transformation protocols were standardized for the black gram (Vigna mungo L.) cv. VBN 3, an important grain legume. An efficient regeneration protocol was developed using 1-week-old de-embryonated cotyledon as explants. The maximum frequency of shoot regeneration (72.6%), multiple shoot induction (6.0 ± 0.3) and shoot elongation was achieved on MS medium supplemented with 3.0 mg/l 6-benzyl amino purine (BAP) after 4 weeks of culture. Up to 65% of the regenerated shoots were rooted on MS medium containing 0.

The present study was carried out to monitor somaclonal variations of banana cv. Ney Poovan (AB) during micropropagation using random amplified polymorphic DNA (RAPD). Sword suckers were used as explants and regenerated on MS media supplemented with BAP alone, BAP + Kinetin and TDZ at varying levels of concentrations. The regenerated shoots were again subcultured on MS medium fortified with plant growth regulators at different concentrations. Rooting was obtained on ½ MS medium supplemented with 2.00 mg/l IBA and 2 g/l of activated charcoal.

Micropropagation is a highly sought after technique in the commercial production of orchid plants. This has the advantage of providing large number of plants in a short period of time. But a major constrain in the in vitro propagation technique is the somaclonal variation. It is important to produce true to type planting materials especially in case of the hybrids as they are more prone to variations. The true to type nature of the micropropagated plantlets can be confirmed by genetic fidelity analysis.

An in vitro protocol for plant regeneration through organogenesis was established for Picrorhiza kurroa Royle Ex. Benth (Scrophulairaceae). The plant is a principle source of glycoside that is Picroside-I, Picroside-II and Kutkoside. Callus was induced on Murashige and Skoog basal medium supplemented with cytokinin (Kn or BA) and auxin (2,4-D/IBA/NAA) using leaf, petiole and stem explants. Shoots were regenerated on MS media with BA (1.5 to 2.5 mgl-1 ) combined with auxin (IBA or NAA) in the range of 0.5 to 2.5 mgl-1 . Rooting was initiated with full or half media supplemented with 0.

Coconut has a wide range of applications from culinary to industrial areas. Loss of coconut trees due to the devastating effect of deadly diseases and the natural aging of the plants demand rejuvenation of orchards with elite seedlings. A conventional breeding program is not sufficient to meet the growing need for healthy planting materials. However, in vitro culture studies can pave the way for mass multiplication. In this study, the effect of TDZ on sliced mature embryo explants through direct organogenesis of coconut var. Chowghat Orange Dwarf (COD) was investigated.

The aim of the present study was to develop a simplified micropropagation protocol for this species, combining phases of in Vitro propagation and rooting and to assess the genetic stability of in Vitro regenerants using ISSR and flow cytometric analysis. The highest number of roots of all tested treatments was produced on MS medium supplemented with zeatin. Ten randomly chosen in Vitro regenerants were subjected to ISSR and flow cytometric analysis. Thus, the micropropagation method optimized here can be used for highly effective production of true-to-type plants of T.Leontopetaloides.

This study was carried out to assess the propagation of Tacca leontopetaloides (Taccaceae) in Vitro using seed embryos. The seeds were cultured on Murashige & Skoog basal media augmented with various concentrations of single or combined growth hormones to obtain a suitable protocol for plantlet regeneration. Seed embryo germination of 57% was obtained, as against 20% for germination under conventional sowing. The best shoot proliferation was obtained using media fortified with 0.1 mg/L 6-benzyl amino purine and 0.

A protocol for direct plantlet regeneration from aerial stem explants of ginger (Zingiber officinale Rosc.) has been developed. Adventitious shoots and roots were regenerated from in planta aerial stem explants of 2 ginger varieties viz. var. ‘Jamaica’ and var. ‘Varada’. Maximum shoot and root regeneration were observed in the cultures containing TDZ and IBA (1:1; 1:0.1 mg L-1) in both varieties.
