The alumina refining industry is one of a number of global industries that produce process waters containing mercury. The research program detailed in this dissertation focuses on processes for removing mercury from Bayer digestion condensate streams. Bayer refinery digestion condensate is chemically unique because it exhibits high pH and is chemically reducing, thermodynamically favouring the presence of elemental mercury.

This research develops a rich theoretical and empirical understanding of embeddedness by way of three dimensions (structural, relational and cultural), and investigates its implications for actors’ value co-creation behaviours and value-in-context outcomes. In addition, the research refines co-creation perspectives by examining a central boundary condition of co-creation processes. The data was collected in an online service environment and was analysed using structural equation modelling techniques.

The aims of this thesis are informed by the limitations of existing research identified above. The overall objective of the thesis is to develop and test a framework of accountant identities (accountant subtypes) inferred from stereotypical perceptions and to identify the attributes that underlie the subtypes. The initial construction of the framework is based on the accountant stereotypes identified in existing research. The framework is constructed from two dimensions based on role (tasks performed by accountants) and character (perceived personality traits).

The X-ray structure of mistletoe lectin I (MLI), a type-II ribosome-inactivatingprotein (RIP), cocrystallized with galactose is described. The model was refined at 3.0 A ˚ resolution to an R-factor of 19.9% using21 899 reflections, with Rfree 24.0%. MLI forms a homodimer (A–B)2in the crystal, as it does in solution at high concentration. The dimer is formed through contacts between the N-terminal domains of two B-chains involvingweak polar and non-polar interactions.

The three-dimensional crystal structure of recombinant annexin Gh1 fromGossypium hirsutum(cotton fibre) has been determined and refined to the finalR-factor of 0.219 at the resolution of 2.1 A ˚ . The solution structural models of AI and AII have been determined in dimethylsulfoxide using NOE distance and 3 JHNHacoupling constants. Finally, theAI family of models resulting from restrained energy minimization (REM) refinement, exhibits pairwise rmsd values for the family ensemble 0.26 ± 0.13 A ˚,1.05 ± 0.23 A ˚, for backbone and heavy atoms, respectively, and the distance penalty function is calculated at 0.

HIV-1 protease (PR) and two drug-resistant variants – PR with the V82A mutation (PRV82A) and PR with the I84V mutation (PRI84V) – were studied using reduced peptide analogs of five natural cleavage sites (CA-p2, p2-NC, p6 pol -PR, p1-p6 and NC-p1) to understand the structural and kine-tic changes. The common drug-resistant mutations V82A and I84V alter residues forming the substrate-binding site. Eight crystal structures were refined at resolutions of 1.10–1.60 A ˚ .

The structure of the complex between the minor groove binder netropsin and d(GGCCAATTGG) was determined via single-crystal X-ray tech-niques. The structure was refined to completion usingrefmac5.1.24, result-ing in a residual R-factor of 20.0% (including 68 water molecules). Using crystal engineering and cryocooling techniques, the resolution could be enhanced to 1.75 A ˚ , resulting in an unambiguous determination of the drug conformation and orientation.

The crystal structure of a subtilisin-like serine proteinase from the psychro-trophic marine bacterium, Vibriosp. PA-44, was solved by means of molecular replacement and refined at 1.84 A˚ . This is the first structure of a cold-adapted subtilase to be determined and its elucidation facilitates examination of the molecular principles underlying temperature adaptation in enzymes.

The crystal structure of the complex between human immunodeficiency virus type 1 (HIV-1) protease and a peptidomimetic inhibitor of ethyleneamine type has been refined toRfactor of 0.178 with diffraction limit 2.5 A ˚ .The peptidomimetic inhibitor Boc-Phe-Y[CH2CH2 NH]-Phe-Glu-Phe-NH2 (denoted here as OE) contains the ethylene-amine replacement of the scissilepeptidebond. The inhibitor lacks the hydroxyl group which is believed to mimic tetra-hedral transition state of proteolytic reaction and thus is suspected to be necessary for good properties of peptido-mimetic HIV-1 protease inhibitors.

A phylogenetic analysis of more than 350 multicopper oxidases (MCOs) from fungi, insects, plants, and bacteria provided the basis for a refined classification of this enzyme family into laccasessensu stricto (basidiomyc-etous and ascomycetous), insect laccases, fungal pigment MCOs, fungal ferroxidases, ascorbate oxidases, plant laccase-like MCOs, and bilirubin oxidases.

Here we report X-ray structures of the chitinase catalytic domain from wild-type (apo,as well as with chloride ions bound) and a Glu234Ala mutant enzyme, solved by molecular replacement and refined at 1.53, 1.8 and 1.7 A˚ resolution, respectively.

A tri-functional monomeric primase–polymerase domain encoded by the plasmid pIT3 fromSulfolobus solfataricusstrain IT3 was identified using a structural–functional approach. The N-terminal domain of the pIT3 repli-cation protein encompassing residues 31–245 (i.e. Rep245) was modeled onto the crystallographic structure of the bifunctional primase–polymerase domain of the archaeal plasmid pRN1 and refined by molecular dynamics in solution.

In this study, the crystal structure of a class Cb-lactamase from a psychro-philic organism,Pseudomonas fluorescens, has been refined to 2.2 A ˚ resolu-tion. It is one of the few solved crystal structures of psychrophilic proteins. The structure was compared with those of homologous mesophilic enzymes and of another, modeled, psychrophilic protein.
