Residual protection

This study examined the phenomenon of residual protection by suspending the operation of established ICCP systems in two wharves located in southern Victoria and monitoring the effect on the passivity of the steel via electrochemical testing. The results of the study confirm that the steel in selected elements within these structures remained passive once the ICCP system was interrupted. The primary factor in determining which elements remained passive, and the duration of passivity was the amount of charge injected into the element over the life of the ICCP system.

The signaling properties of the gasotransmitter molecule hydrogen sulfide (H2S), which is endogenously generated in plant cells, are mainly observed during persulfidation, a protein post-translational modification (PTM) that affects redox-sensitive cysteine residues. There is growing experimental evidence that H2S in higher plants may function as a mechanism of response to environmental stress conditions.

The involvement of the lysine residue present at the active site of Ehrlich ascites carcinoma (EAC) cell glyceraldehyde-3-phosphate dehydrogenase (Gra3P DH) was investigated by using the lysine specific reagents trinitrobenzenesulfonic acid (TNBS) and pyridoxal phosphate (PP). Both TNBS and PP inactivated EAC cell Gra3P DH with pseudo-first-order kinetics with the rate dependent on modifier concentration. Kinetic analysis, including a Tsou plot, indicated that both TNBS and PP apparently react with one lysine residue per enzyme molecule.

Biosynthesis ofmucin-typeO-glycans is initiated by a family of UDP-GalNAc:polypeptide N-acetylgalactosaminyl-transferases, which contain several conserved cysteine resi-dues among the isozymes. We found that a cysteine-specific reagent, p-chloromercuriphenylsulfonic acid (PCMPS), irreversibly inhibited one of the isozymes (GalNAc-T1). Presence of either UDP-GalNAc or UDP during PCMPS treatment protected GalNAc-T1 from inactivation, to the same extent.

Neutralizing and protective monoclonal antibodies (mAbs) were used to fine-map the highlyconserved hemagglutinin noose epitope (H379–410, HNE) of themeasles virus. Short peptides mimicking this epitope were previouslyshown to induce virus-neutralizing antibodies [El Kasmi et al. (2000) J. Gen. Virol.81, 729–735]. The epitope contains three cys-teine residues, two of which (Cys386 and Cys394) form a disulfide bridge critical for antibodybinding.

Plasminogen activator inhibitor-1 (PAI-1) belongs to the serpin family of serine proteinase inhibitors. Serpins inhibit their target proteinases by an ester bond being formed between the active site serine of the proteinase and the P1 residue of the reactive centre loop (RCL) of the serpin, fol-lowed by insertion of the RCL intob-sheet A of the serpin. Concomitantly, there are conformational changes in the flexible joint region lateral tob-sheet A.

Silk fibroin ofBombyx moriis secreted from the posterior silkgland (PSG) as a2.3-MDa elementaryunit, consistingof six sets of a disulfide-linked heavy chain (H-chain)–light chain (L-chain) heterodimer and one molecule of fibrohex-amerin (fhx)/P25. Fhx/P25, a glycoprotein, associates non-covalently with the H–L heterodimers. The elementary unit was found and purified from the endoplasmic reticulum (ER) extract of PSG cells.
