In this study a novel protein isolated from Trichosanthes dioica of Cucurbitaceae family which upon testing exhibited ribosome inactivating property was tested for anticancer properties. This may pave the way for exploration of these proteins for potential use as anti-cancer agents in future.

Bougainvillea antiviral protein (BAP) is one among a class of the ribosomal inactivating proteins isolated from Bougainvillea spectabilis willd. Truncated version of the BAP gene was cloned and expressed in a prokaryotic vector to abolish its cytotoxicity. RNA was isolated from mature leaves of Bougainvillea and the full length cDNA was amplified by reverse transcription-PCR using template mRNA. This full length cDNA of size 756 bp was amplified using the proofreading polymerase (Q5 polymerase) and end to end gene specific primers for removal of C-terminal, the amplicon was cloned in pJET1.

Lamjapin,anoveltypeIribosome-inactivating protein, has been isolated from kelp (Laminaria japonicaA), a marine alga. This protein has been extensively purified through multiple chromatography columns. With a molecular mass of36 kDa, lamjapin is slightly larger than the other known single-chain ribosome-inactivating proteins from the higher plants. The X-ray structure of mistletoe lectin I (MLI), a type-II ribosome-inactivatingprotein (RIP), cocrystallized with galactose is described. The model was refined at 3.0 A ˚ resolution to an R-factor of 19.9% using21 899 reflections, with Rfree 24.0%. MLI forms a homodimer (A–B)2in the crystal, as it does in solution at high concentration. The dimer is formed through contacts between the N-terminal domains of two B-chains involvingweak polar and non-polar interactions.

Abstract X-ray crystal structure determination of agglutinin from abrus precatorius in Taiwan is presented. The crystal structure of agglutinin, a type II ribosome-inactivating protein (RIP) from the seeds of Abrus precatorius in Taiwan, has been determined from a novel crystalline form by the molecular replacement method using the coordinates of abrin-a as the template. The structure has space group P41212 with Z = 8, and been refined at 2.6 Å to R-factor of 20.4%. The rootmean-square deviations of bond lengths and angles from the standard values are 0.009 Å and 1.3°.
