The recombinant polymerases of human influenza virus A/PR/8/34 (H1N1) (PR8) and avian influenza virus A/Turkey/England/1969 (H3N2) (TE) using anti-FLAG M2 affinity resin. After confirming trimeric complexes, enzymatic properties of recombinant polymerases were characterized, including model viral RNA binding, in vitro transcription assays, and cap-snatching activity upon addition of cap1-70 mer vRNA as substrate. Taken together we conclude that 293T cells are a suitable expression system for sufficient amount isolation of functional recombinant influenza viral polymerases with 95% of purity.

The nonphotosynthetic mutant ofArabidopsis hcf152is impaired in the processing of the chloroplast polycistronic transcript, psbB-psbT-psbH-petB-petD, resulting in non-production of the essential photosynthetic cytochromeb6 f complex. The nucleus-encodedHCF152gene was identified to encode a pentatricopeptide repeat (PPR) protein com-posed primarily of 12 PPR motifs, similar to other proteins of this family that were identified in mutants defected in chloroplast gene expression.

In our attempt to obtain further information on the replication mechanism of the hepatitis C virus (HCV), we have studied the role of sequences at the 3¢-end of HCV minus-strand RNA in the initiation of synthesis of the viral genome by viral RNA-dependent RNA polymerase (RdRp). In this report, we investigated the template and binding properties of mutated and deleted RNA fragments of the 3¢-end of the minus-strand HCV RNA in the presence of viral polymerase.

Hfq proteins are common in many species of enterobacteria, where they participate in RNA folding and translational regulation through pairing of small RNAs and messenger RNAs. Hfq proteins share the distinctive Sm fold, and form ring-shaped structures similar to those of the Sm⁄Lsm pro-teins regulating mRNA turnover in eukaryotes.
