Would you be enticed by an ad for a coat that is waterproof, stretchable, washable, and permanent-press, that automatically repairs small cuts, rips, and burns? How about one that’s guaranteed to last a lifetime? Sounds too good to be true, but you already have such a coat-your skin. The skin and its derivatives (sweat and oil glands, hairs, and nails) make up a complex set of organs that serves several functions, mostly protective. Together, these organs form the integumentary system.

Venoms from crotalid and viperid snakes contain several peptide inhibitors which regulate the proteolytic activities of their snake-venom metalloproteinases (SVMPs) in a reversible manner under physiological conditions. In this report, we describe the high-resolution crystal structures of a SVMP, TM-3, from Taiwan habu (Trimeresurus mucrosquamatus) cocrystallized with the endogenous inhibitors pyroGlu-Asn-Trp (pENW), pyroGlu-Gln-Trp (pEQW) or pyroGlu-Lys-Trp (pEKW).

Angiotensin converting enzyme (ACE) was already discovered in insects in 1994, but its physiological role is still enigmatic. We have addressed this problem by purifying four new ACE substrates from the ovaries of the grey fleshfly, Neobellieria bullata. Their primary structures were identified as NKLKPSQWISLSD (Neb-ODAIF-11)13), NKLKPSQWI (Neb-ODAIF-11)9), SLKPSNWLTPSE (Neb-ODAIF-2) and LEQIYHL. Database analysis showed significant homology with amino acid sequence stretches as present in the N-terminal part of several fly yolk proteins....

Antifreeze proteins (AFPs) are produced by several cold-water fish species. They depress physiological freezing temperatures by inhibiting growth of ice crystals and, in so doing, permit the survival of these fish in seawater cooler than their normal freezing temperatures. The type II AFP from rainbow smelt (Osmerus mordax), which is a member of the C-type lectin superfamily, was characterized in terms of its Ca2+-binding quaternary structure and the role of its single N-linked oligosaccharide....

Oviparously developing embryos of the brine shrimp, Artemia franciscana, synthesize abundant quantities of a small heat shock/a-crystallin protein, termed p26. Wild-type p26 functions as a molecular chaperone in vitro and is thought to help encysted Artemia embryos survive severe physiological stress encountered during diapause and anoxia. Full-length and truncated p26 cDNA derivatives were generated by PCR amplification of p26-3-6-3, then cloned in either pET21(+) or pRSETC and expressed in Escherichia coli BL21(DE3)....

The adenosine A2a receptor belongs to the seven trans-membrane helix G-protein-coupled receptor family, is abundant in striatum, vasculature and platelets and is involved in several physiological processes such as blood pressure regulation and protection of cells during anoxia. For structural and biophysical studies we have expressed the human adenosine A2areceptor (hA2aR) at high levels inserted into the Escherichia coliinner membrane, and established a puri®cation scheme.

Plant lipoxygenases (LOXs) are a class of dioxygenases which display diverse functions in several physiological processes such as growth, development and response to biotic and abiotic stresses.Even though LOXs have been characterized from several plant species, the physiological role of seed LOXs is still unclear. With the aim to better clarify the occurrence of LOXs and their influence on hazelnut seed quality, we carried out the biochemical and molecular characterization of the main LOX isoforms expressed during seed development....

To assess the expression and physiological role of the mitochondrial NAD + -independent lactate dehydrogenase (iLDH) inEuglena gracilis, cells were grown with different carbon sources, and theD-andL-iLDHactivities and several key metabolic intermediates were examined. iLDH activity was significant throughout the growth period, increasing by three- to fourfold from latency to the stationary phase. Intracellular levels ofD-andL-lactate were high (5–40 mM) fromthe start of the culture and increased (20–80 mM)when the stationaryphasewas entered....

NADHoxidase fromThermus thermophilusis a homodimer with anunknownphysiological function. As is typical for an enzyme isolated froma thermophile, the catalytic rate, kcat,is low at low temperatures and increases with temperature, achieving an optimum at the physiological temperature of the organism,i.e. at 70CforT. thermophilus.Atlow temperatures, thekcatof several enzymes from thermophilic and mesophilic organisms can be increased by chaotropic agents.

The formation of amyloid fibrils is associated with various human medical disorders of unrelated origin. Recent research indicates that self-assembled amyloid fibrils are also involved in physiological processes in several micro-organisms. Yet, the molecular basis for the recognition and self-assembly processes mediating the formation of such structures from their soluble protein precursors is not fully understood.

In vertebrates, multiple microsomal retinol dehydrogenases are involved in reversible retinol⁄retinal interconversion, thereby controlling retinoid meta-bolism and retinoic acid availability. The physiologic functions of these enzymes are not, however, fully understood, as each vertebrate form has several, usually overlapping, biochemical roles.

The endocannabinoid system is a conserved system involved in the modula-tion of several physiologic processes, from the activity of the central ner-vous system to reproduction. Type 1 cannabinoid receptor (CNR1) cDNA was cloned from the brain and testis of the anuran amphibian, the frog Rana esculenta.

The mammalian brain contains unusually high levels ofd-serine, ad-amino acid previously thought to be restricted to some bacteria and insects. In the last few years, studies from several groups have demonstrated that d-serine is a physiological co-agonist of the N-methyl d-aspartate (NMDA) type of glutamate receptor – a key excitatory neurotransmitter receptor in the brain.

This concise volume, edited by two of today’s leading pain clinician-scientists, represents the culmination of several forces. First and foremost is the recognition that the knowledge and skills supporting current medical management of pain have grown sufficiently large that this field has become a discipline in its own right. Accordingly, candidates who meet the requirements of the American Board of Anesthesiology may now become board-certified in Pain Management and achieve diplomate status just as their colleagues in other areas have done for years.

For many years obesity in rodents has been known to be caused by a number of distinct mutations distributed through the genome. Most of these singlegene mutations cause both hyperphagia and diminished energy expenditure, suggesting a physiologic link between these two parameters of energy homeostasis. Identification of the ob gene mutation in genetically obese (ob/ob) mice represented a major breakthrough in the field. The ob/ob mouse develops severe obesity, insulin resistance, and hyperphagia, as well as efficient metabolism (e.g.
