Viêm gan tự miễn (VGTM) là bệnh thường ít được nghĩ tới ở tình huống có tăng men gan, có dấu ấn huyết thanh của virus viêm gan (+) và xét nghiệm tự kháng thể ANA (-). Bệnh có thể gây xơ gan và thậm chí tử vong nếu không được chẩn đoán và điều trị. Tại bệnh viện Chợ Rẫy chúng tôi ghi nhận một số ca VGTM không điển hình với các đặc điểm nêu trên, đáp ứng tốt với với điều trị ức chế miễn dịch.

The main component of cerebral amyloid angiopathy (CAA) in Alzheimer’s disease is the amyloid-b protein (Ab), a 4-kDa polypeptide derived from the b-amyloid protein precursor (APP). The accumulation of Ab in the basement membrane has been implicated in the degeneration of adjacent vascular smooth muscle cells (VSMC). However, the mechanism of Ab toxicity is still unclear. In this study, we examined the effect of substrate-bound Ab on VSMC in culture. The use of substrate-bound proteins in cell culture mimics presentation of the proteins to cells as if bound to the basement membrane.

In this study, we isolated a 25-kDa novel snake venom protein, designated ablomin, from the venom of the Japanese Mamushi snake (Agkistrodon blomhoffi). The amino-acid sequence of this protein was determined by peptide sequencing and cDNA cloning. The deduced sequence showed high similarity to helothermine from the Mexican beaded lizard (Heloderma horridum horridum), which blocks voltage-gated calcium and potassium channels, and ryanodine receptors. The effect of a-tocopheryl hemisuccinate (TS) on lipopolysaccharide (LPS)/interferon-c (IFN)-induced nitric oxide production in rat vascular smooth muscle cells (VSMC) was examined. The LPS/IFN-induced NO production was enhanced by TS but not by the other a-tocopherol (a-T) derivatives a-tocopheryl acetate (TA) and a-tocopheryl nicotinate (TN), or a-T itself. a-T, TA and TN inhibited the enhancement by TS of LPS/IFNinduced NO production.

Two neuropeptides have been isolated and identi®ed from the secretions of the skin glands of the Stony Creek Frog Litoria lesueuri. The ®rst of these, the known neuropeptide caerulein 1.1, is a common constituent of anuran skin secretions, and has the sequence pEQY(SO3 )TGWMDF-NH2 . This neuropeptide is smooth muscle active, an anal-gaesic more potent thanmorphine and is also thought to be a hormone.

ADP-ribosylation has been coupled to intracellular events associated with smooth muscle cell vasoreactivity, cytoskel-etal integrity and free radical damage. Additionally, there is evidence that ADP-ribosylation is required for smooth muscle cell proliferation. Activation of a calcium-sensing receptor (Ca-SR) leads to increased intracellular calcium concentration and altered cellular activities. The expression of Ca-SR has been iden-tified in both nonexcitable and excitable cells, including neurons and smooth muscle cells. Whether Ca-SR was expressed and functioning in cardiac myocytes remained unclear. In this study, we have characterized the influence of hypoxia on the expression of hydroxylases crucially involved in col-lagen fiber formation, such as prolyl-4-hydroxylases (Ph4) and procollagen lysyl-hydroxylases (PLOD). Using the rat vascular smooth muscle cell line A7r5, we found that an hypoxic atmosphere caused a characteristic time-dependent five- to 12-fold up-regulation of the mRNAs of the two P4h a-subunits [aI (P4ha1) andaII (P4ha2)] and of two lysyl-hydroxylases (PLOD1 and PLOD2).

The proliferation of smooth muscle cells (SMC) is a key event in the development of atherosclerosis. In addition to growth factors or cytokines, we have shown previously that n-3 polyunsaturated fatty acids (PUFAs) act in opposition to n-6 PUFAs by modulating various steps of the inflam-matory process. Connective tissue growth factor (CTGF/CCN2) is an immediate early gene-encoded polypeptide modulating cell growth and collagen synthesis. The importance of CTGF/ CCN2 function is highlighted by its disregulation in fibrotic disorders. In this study, we investigated the regulation and signaling pathways that are required for various stimuli of intracellular signaling events to induce the expression of the endogenous CTGF/CCN2gene in smooth muscle cells.

We used differential scanning calorimetry (DSC) and circular dichroism (CD) to investigate thermal unfolding of recombinant fibroblast isoforms ofa-tropomyosin (Tm) in comparison with that of smooth muscle Tm. These two nonmuscle Tm isoforms 5a and 5b differ internally only by exons 6b⁄6a, and they both differ from smooth muscle Tm by the N-terminal exon 1b which replaces the muscle-specific exons 1a and 2a.
