The multisubstrate nucleoside kinase of Drosophila melanogaster(Dm-dNK) can be expressed in human solid tumor cells and its unique enzymatic properties makes this enzyme a suicide gene candidate. In the present study,Dm-dNK was stably expressed in the CCRF-CEM and H9 T-lymphoblastoid cell lines. The expressed enzyme was localized to the cell nucleus and the enzyme retained its activity.

Human cytochrome P450 3A4 forms a series of minor testosterone hydroxylation products in addition to 6b-hy-droxytestosterone, the major product. Microfibrils and elastin are major constituents of elastic fibers, the assembly of which is dictated by multimolecular interactions.Microfibril-associated glycoprotein-1 (MAGP-1) is a microfibrillar component that interacts with the sol-uble elastin precursor, tropoelastin. We describe here the adaptation of a solid-phase binding assay that defines the effect of divalent cations on the interactions between MAGP-1 and tropoelastin. Using this assay, a strong cal-cium-dependent interaction was demonstrated, with a dis-sociation constant of 2.8 ± 0.3 nM, which fits a single-site binding model.

We utilized plasmon-waveguide resonance (PWR) spectroscopy to follow the effects of sphingomyelin, cholesterol and zinc ions on the binding and aggregation of the amyloidbpeptide1)40 in lipid bilayers. With a dioleoyl-phosphatidylcholine (DOPC) bilayer, peptide binding was observed, but no aggregation occurred over a period of 15 h.

The chaperone calreticulin has been suggested to function as a C1q and collectin receptor. The interaction of calreticulin with mannan-binding lectin (MBL) was investigated by solid-phase binding assays.
