The in vitro raised plantlets were successfully acclimatized under ex vitro conditions and grew well in the glasshouse. All the regenerated plantlets appeared normal with respect to morphological characteristics with 100% survival rate. Somaclonal variation in tissue culture is a common phenomenon which makes it mandatory to check for genetic stability of plants. The genetic variation in the in vitro raised plants was confirmed using Inter Simple Sequence Repeats(ISSR) markers. Comparison of the bands with the mother plant revealed that the genetic variation frequency reached 6.

Genetic uniformity of in vitro raised plants is a prerequisite for production of quality planting material of banana. Embryogenic cultures were induced from immature male flower buds of two important cultivars of banana, ‘Rasthali’ (Silk-Musa AAB) and ‘Grand Naine’ (Cavendish-Musa AAA). Cell suspensions were established and regenerated plants were evaluated for their genetic stability using 10 inter simple sequence repeat markers.

The experiment was taken up to elicit the information on yield attributes and yield of different ginger (Zingiber officinale Rosc.) somaclones. Ten somaclones of ginger were evaluated in RBD with three replications during 2015-18 at College of Horticulture, Kerala Agricultural University, Thrissur district, Kerala. The growth performance of ten somaclones indicated significant variation at all the stages of crop growth. The somaclones SE 86102 (16.00 days) and SE 8626 (16.33 days) registered least number of days for sprouting.

Thirteen groups of somaclones (seven in Maran and six in Rio-de-Janeiro) were evaluated using Random Amplified Polymorphic DNA (RAPD). RAPD analysis using twelve selected primers produced 129 amplicons, 44 were polymorphic with an average of 3.66 polymorphic bands / primer and a polymorphism percentage of 34.10. The dendrograms generated based on RAPD profiles grouped the somaclones into two separate clusters. RAPD marker system showed that somaclones derived from cultivar Maran exhibited more variability than Rio-de-Janeiro.

In this study, RAPD fingerprinting has been carried out for variants selected from orchards growing with micropropagated Robusta banana (Musa spp. ‘AAA’) plants. After isolation of quality DNA and preliminary screening of random primers, fourteen arbitrary (10-mers) primers showing intense unambiguous and reproducible PCR amplification have been selected for study. Among them OPA-19 and OPC-03 showed only monomorphic bands while OPA-02, OPA-09, OPA-13, OPA-14, OPB-6, OPB-15, OPC-01, OPD-10, OPF-04 and OPF-12 primers showed polymorphic bands.

Assessment of genetic polymorphism in sugarcane somaclones by using random amplified polymorphic DNA
DNA was extracted from plants and RAPD analysis was performed for fourteen treatments with two random oligonucleotide primers (OPA-01 and OPB-11). Among fourteen treatments, plants derived from the treatments T9, T11, and T12 produced polymorphism T12 was found to exhibit maximum polymorphism at 1.0 µM EMS in 2009T5. The clone, 2009T5 produced more variation than 2008T42. Sodium nitrite did not produce any variation.

The present study was carried out to monitor somaclonal variations of banana cv. Ney Poovan (AB) during micropropagation using random amplified polymorphic DNA (RAPD). Sword suckers were used as explants and regenerated on MS media supplemented with BAP alone, BAP + Kinetin and TDZ at varying levels of concentrations. The regenerated shoots were again subcultured on MS medium fortified with plant growth regulators at different concentrations. Rooting was obtained on ½ MS medium supplemented with 2.00 mg/l IBA and 2 g/l of activated charcoal.

Present investigation was carried out to screen two promising sugarcane varieties CoN 13073 and CoN 09072 for salinity tolerance through callus culture. Various physiological and morphological parameters was studied, maximum shoot length was observed at 2.0 % NaCl concentration treated to callus at 65 days of culturing in variety CoN 13073, similar trend was observed for root length and leaf number per plant in variety CoN 13073 at 2.0 % NaCl concentration..

Micropropagation is a highly sought after technique in the commercial production of orchid plants. This has the advantage of providing large number of plants in a short period of time. But a major constrain in the in vitro propagation technique is the somaclonal variation. It is important to produce true to type planting materials especially in case of the hybrids as they are more prone to variations. The true to type nature of the micropropagated plantlets can be confirmed by genetic fidelity analysis.

Tissue culture regenerated materials of Musa L. spp. (banana and plantain) along with their somaclonal variants were assessed for karyotype abnormalities to be used as indicators for rapid cytological marking. The results show that although slight variations were noted in karyomorphometric characters, like chromosome number, arm length, total chromosome length, and total chromosome volume, these were not significant (P ≤ 0.05).
