Twenty-four branched maltodextrins were synthesized from eight starches using three thermostable microbial glycogen branching enzymes. The maltodextrins have a degree of branching (DB) ranging from 5 % to 13 %. This range of products allows us to explore the effect of DB on the digestibility, which was quantified under conditions that mimic the digestion process in the small intestine.

This study was aimed to screen and isolate thermophilic fungi from three selected compost wastes sites (Palm oil mill, Wood chip piles and Abattoir dump), and identify potential producers of lipases, cellulases and proteases respectively. The thermophilic fungi were obtained from an equal depth of one meter in all the three collection sites with a temperature range of 40 to 45OC and later cultured at 70 OC. F

The selectivity and the thermostability of EndoBI-1 and EndoBI-2 suggest that these enzymes may be useful for many scientific and industrial applications. In this study, the growing numbers of homologous sequences in different databases were exploited in a comparative approach to investigate structural properties of EndoBI-1 and EndoBI-2 enzymes. Moreover, the complete and partial homology models of these two enzymes were generated and evaluated.

The present study describes the simultaneous expression of thermostable industrial alpha (α) and beta (β) amylase enzymes that have been used widely in starch industry. Genomic DNA of Bacillus stearothermophilus DSM 22 strain for α amylase and, Thermoanaerobacterium (Clostridium) thermosulfurogenes DSM 2229 strain for β amylase were used as gene sources. Both genes were ligated into pETDuet-1 expression vector separately and resulting recombinant vectors were transformed into Escherichia coli BL21 competent cells by electroporation.

Peptidases (EC 3.4) consist of a large group of hydrolytic enzymes that catalyze the hydrolysis of proteins accounting for approximately 65% of the total worldwide enzyme production. Peptidases from thermophilic fungi have adaptations to high temperature that makes them adequate for biotechnological application. In the present study, we profiled the genomes of heat-tolerant fungi and phylogenetically related mesophilic species for genes encoding for peptidases and their putative adaptations for thermostability.

The thermophile bacteria that came from hot spring, generally, it is thermostable, which it can produce enzyme. For example, there are protease, lipase, and amylase. Amylase produced by bacteria or bacteria amylase is an enzyme that can hydrolyze starch to sugar. The thermostable of amylase resist to high temperature and pH. It has been used on many industrial productions like foods, fermentation, textile, alcohol, paper, pharmaceutical and detergent.

Lipases are enzymes that catalyze the transformation reactions of triglycerides in to fatty acids and glycerol. They can be produced by animals, plants and microorganisms. This article presents the isolation of lipase-producing bacteria from soil samples collected from sites contaminated with waste from palm oil production in the littoral region of Cameroon. These permit to isolate a multitude of bacteria capable of producing lipase from Rhodamine B-agar olive oil culture medium.

In this study, an extracellular alkali-thermostable phytase producing bacteria, Bacillus subtilis B.S.46, were isolated and molecularly identified using 16S rRNA sequencing. Response surface methodology was applied to study the interaction effects of assay conditions to obtain optimum value for maximizing phytase activity. The optimization resulted in 137% (4.627 U/mL) increase in phytase activity under optimum condition (56.5 C, pH 7.30 and 2.05 mM sodium phytate). The enzyme also showed 60–73% of maximum activity at wide ranges of temperature (47–68 C), pH (6.3–8.

The major cellulase secreted by the filamentous fungus Trichoderma reeseiis cellobiohydrolase Cel7A. Its three-dimensional structure has been solved and various mutant enzymes produced. In order to study the potential use of T. reeseiCel7A in the alkaline pHrange, the thermal sta-bilityofCel7Awas studiedas a functionof pHwith thewild-type and two mutant enzymes using different spectroscopic methods. Tryptophan fluorescence and CD measurements of the wild-type enzyme show an optimal thermostability between pH3.5–5.

We report herein the first molecular characterization of 5¢-deoxy-5¢-methyl-thio-adenosine phosphorylase II from Sulfolobus solfataricus(SsMTAPII). The isolated gene of SsMTAPII was overexpressed inEscherichia coli BL21. Purified recombinant SsMTAPII is a homohexamer of 180 kDa with an extremely low Km(0.7lm) for 5¢-deoxy-5¢-methylthioadenosine. The enzyme is highly thermophilic with an optimum temperature of 120C and extremely thermostable with an apparent Tmof 112C that increases in the presence of substrates....
