The plastid transcription kinase (PTK), a component of the major RNA polymerase complex from mustard chloroplasts, has been implicated in redox-mediated regulation of plastid gene expression. A cloning strategy to define the PTK gene(s) resulted in the isolation of a full-length cDNA for a protein with overall high homology with the a subunit of cytosolic casein kinase (CK2) that contained an N-terminal extension for a putative plastid transit peptide.

The temperature-induced structural transitions of the full length Alzheimer amyloid b-peptide [Ab(1–40) peptide] and fragments of it were studied using CD and 1 H NMR spectroscopy. The full length peptide undergoes an overall transition from a state with a prominent population of left-handed 31 (polyproline II; PII)-helix at 0C to a random coil state at 60C, with an averageDH of 6.8 ± 1.4 kJÆmol )1 per residue, obtained by fitting a Zimm–Bragg model to the CD data.

The mechanism of inhibition of yeast F0F1-ATPase by its naturally occurring protein inhibitor (IF1) was investi-gated in submitochondrial particles by studying the IF1-mediatedATPase inhibition in the presence andabsence of a protonmotive force. In order to evaluate the role of 3(S)-hydroxyproline [3(S)-Hyp] in the triple-helical structure, we produced a series of model peptides with nine tripeptide units including 0–9 3(S)-hydroxyproline residues. The sequences are H-(Gly-Pro-4(R)Hyp) l -(Gly-3(S)Hyp-4(R)Hyp)m-(Gly-Pro-4(R)Hyp)n -OH, where (l, m, n) = (9, 0, 0), (4, 1, 4), (3, 2, 4), (3, 3, 3), (1, 7, 1) and (0, 9, 0). All peptides showed triple-helical CD spectra at room temperature and thermal transition curves.

Plastid envelope DNA-binding protein (PEND) is a DNA-binding protein with a chloroplast basic region-zipper domain at its N-terminus and a transmembrane domain at its C-terminus. The N-termini of the NADPH : protochlorophyllide oxidoreductase (POR) proteins A and B from barley and POR from pea were determined by acet-ylation of the proteins and selective isolation of the N-terminal peptides for mass spectrometry de novosequence analysis. We show that the cleav-age sites between the transit peptides and the three mature POR proteins are homologous.
