Physics based process modeling using finite element method (FEM) has been integrated with the soft computing techniques like artificial neural networks (ANN) and genetic algorithm (GA) to improve prediction accuracy of the model with less dependency on the experimental data.

The chaperone calreticulin is a highly conserved eukaryotic protein mainly located in the endoplasmic reticulum. It contains a free cysteine SH group but does not form disul-fide-bridged dimers under physiological conditions,indica-ting that the SH group may not be fully accessible in the native protein. Using PAGE,urea gradient gel electro-phoresis,capillary electrophoresis and MS,we show that dimerization through the SH group can be induced by lowering the pH to 5–6,heating,or under conditions that favour partial unfolding such as urea concentrations above 2.

We havemade a comprehensive and quantitative analysis of the lipid composition of caveolae from primary rat fat cells and compared the composition of plasma membrane inside and outside caveolae. We isolated caveolae from purified plasmamembranesusingultrasonication incarbonatebuffer to disrupt the membrane, or extraction with nonionic detergent, followed by density gradient ultracentrifugation. The carbonate-isolated caveolae fraction was further immunopurified using caveolin antibodies.

NF-jB essential modulator (NEMO) plays an essential role in the nuclear factor jB (NF-jB) pathway as a modulator of the two other subunits of the IjB kinase (IKK) complex, i.e. the protein kinases, IKKa and IKKb. Previous reports all envision the IKK complex to be a static entity. Using glycerol-gradient ultracentrifugation, we observed stimulus-dependent dynamic IKK complex assembly.
