Deproteinated cheese whey was used for biomass production of probiotic strain Lactobacillus helveticus MTCC 5463 in a biofermenter. Optimization of growth parameters such as temperature, pH and time of incubation as well as nutrient supplementation of cheese whey was carried out using response surface methodology (RSM). Cheese whey supplemented with 0.95% yeast extract and 0.95% proteose peptone, inoculated with 6% (v/v) active culture of L. helveticus MTCC 5463 and fermented for 24 h at optimized temperature of 40ºC and pH 6.25 yielded 3.25 g/L dry cell biomass and 14.

The addition of glucose toSaccharomyces cerevisiaecells causes reprogramming of gene expression. Glucose is sensed bymembrane receptorsaswell as (so far elusive) intracellular sensing mechanisms. The availability of four yeast strains that display different hexose uptake capacities allowed us to study glucose-induced effects at different glycolytic rates. Rapid glucose responses were observed in all strains able to take up glucose, consistent with intracellular sensing.

During a search for cDNAs encoding plant sterol acyl-transferases, we isolated four full-length cDNAs fromAra-bidopsis thaliana that encode proteins with substantial identity with animal lecithin : cholesterol acyltransferases (LCATs). The expressionof one of these cDNAs,AtLCAT3 (At3g03310), invarious yeast strains resulted in the doubling of the triacylglycerol content.

A fatty-acid-metabolizing enzyme fromArabidopsis thaliana, CYP94C1, belonging to the cytochrome P450 family was cloned and characterized. CYP94C1 was heterologously expressed in aSaccharomyces cerevisiae strain (WAT11) engineered for P450 expression. When recombinant yeast microsomes were incubated with lauric acid (C12:0) for 15 min, one major metabolite was formed.

The assembly status of the cytochromebc1 complex has been analyzed in distinct yeast deletion strains in which genes for one or more of thebc1 subunits were deleted. In all the yeast strains tested, a bc1 sub-complex of approximately 500 kDa was found when the mitochondrial membranes were analyzed by blue native electrophoresis.

Problem statement: Dilute sulphuric acid and enzymatic hydrolysis methods were used for sugar extraction. Xylose and glucose sugars were obtained from corn cobs. Approach: Acid hydrolysis of corn cobs gave higher amount of sugars than enzymatic hydrolysis. Results: The results showed that optimal temperature and time for sugar fermentation were approximately 25°C and 50 h by two yeast strains (S. cerevisiae and P. Stipitis) respectively.
