This work demonstrated the potential of US for mitigating membrane fouling. However, the integration of the US transducers within the membrane system will be a major challenge for the application of US in membrane cleaning. It is more appropriate that US is used as pre-treatment to the membrane system, the transducers can be placed prior to the coagulant feed line. Further work on the determination of the detailed mechanisms associated with enhancing coagulation would enable optimisation of the system with regard to process, cost and energy efficiency.

The main aim of this research is to develop a viable solution to treat and reuse car wash wastewater. To do so, two treatment systems were evaluated. The first treatment system comprised of coagulation-flocculation and ceramic ultrafiltration membrane and reverse osmosis and the second treatment system is enhanced membrane bioreactor (eMBR).

The interaction of blood coagulation factor X and its Gla-containing fragments with negatively charged phospholipid membranes composed of 25 mol% phosphatidylserine (PtdSer) and 75 mol% phosphatidylcholine (PtdCho) was studied by surface plasmon resonance. The binding to 100 mol% PtdCho membranes was negligible. The calcium dependence in the membrane binding was evaluated for intact bovine factor X (factor X) and the fragment containing the Gla-domain and the N-terminal EGF (epidermal growth factor)-like domain, Gla–EGFN, from factor X....

Kininogens are multifunctional proteins found so far mainly in mammals. They carry vasoactive kinins as well as participate in defense, blood coagulation and the acute phase response. In this study, novel kininogens were isolated from Atlantic cod (Gadus morhua L.) and spotted wolffish (Anarhichas minor) by papain-affinity chromatography. The molecular mass of cod kininogen determined by MALDITOF mass spectrometry to be 51.0 kDa and it had pI values of 3.6, 3.9 and 4.4.

Here we have characterized variants of FVIIa mutatedat one or twoof these positions toassess the relative importance of the individual replacements.

The immune coagulant fgl2/fibroleukin has been previously shown to play a pivotal role in the pathogenesis of murine and human fulminant hepatitis and fetal loss syndrome. Constitutive expression of fgl2 transcripts at low levels are seen in cytotoxic T cells, endothelial, intestinal and tropho-blast cells, while specific factors (such as virus and cytokines) are required to induce high levels of fgl2 expression in other cell types including monocytes/macrophages.

Members of the serine protease inhibitor (serpin) superfamily play import-ant roles in the inhibition of serine proteases involved in complex systems. This is evident in the regulation of coagulation serine proteases, especially the central enzyme in this system, thrombin. This review focuses on three serpins which are known to be key players in the regulation of thrombin: antithrombin and heparin cofactor II, which inhibit thrombin in its pro-coagulant role, and protein C inhibitor, which primarily inhibits the throm-bin⁄thrombomodulin complex, where thrombin plays an anticoagulant role. .

Exposure of blood to tissue factor (TF) rapidly initiates the coagulation serine protease cascades. TF is expressed by macrophages and other types of cell within atherosclerotic lesions and plays an important role in throm-bus formation after plaque rupture. Macrophage TF expression is induced by pro-inflammatory stimuli including lipopolysaccharide (LPS), inter-leukin-1band tumor necrosis factor-a. Here we demonstrate that activation of liver X receptors (LXRs) LXRaand LXRbsuppresses TF expression....

Uniquely amongst vitamin K-dependent coagulation proteins, protein C interacts via its Gla domain both with a receptor, the endothelial cell protein C receptor (EPCR), and with phospholipids. We have studied naturally occurring and recombinant protein C Gla domain variants for soluble (s)EPCR binding, cell surface activation to activated protein C (APC) by the thrombin–thrombomodulin complex, and phospholipid dependent factor Va (FVa) inactivation by APC, to establish if these functions are concordant. ...

Procarboxypeptidase U [proCPU, thrombin-activatable fibrinolysis inhib-itor (TAFI), EC 3.4.17.20] belongs to the metallocarboxypeptidase family and is a zymogen found in human plasma. ProCPU has been proposed to be a molecular link between coagulation and fibrinolysis. Upon activation of proCPU, the active enzyme (CPU) rapidly becomes inactive due to its intrinsic instability.

Activation of coagulation factor X (fX) by activated factors IX (fIXa) and VIII (fVIIIa) requires the assembly of the enzyme–cofactor–substrate fIXa– fVIIIa–fX complex on negatively charged phospholipid membranes. Using flow cytometry, we explored formation of the intermediate membrane-bound binary complexes of fIXa, fVIIIa, and fX.

The coagulation factor X activator from Russell’s viper venom (RVV-X) is a heterotrimeric glycoprotein. In this study, its three subunits were cloned and sequenced from the venom gland cDNAs of Daboia siamensis. The deduced heavy chain sequence contained a C-terminal extension with four additional residues to that published previously.

During thrombus formation, thrombin, which is abundantly present at sites of vascular injury, activates platelets in part via autocrine-produced ADP. We investigated the signaling pathways by which thrombin and ADP in synergy induced platelet Ca 2+ elevation and procoagulant activity, and we monitored the consequences for the coagulation process.

Coagulation factor XI (FXI) is the zymogen of a serine protease that, when converted to its active form, contributes to blood coagulation through proteolytic activation of factor IX. FXI deficiency is typically an autosomal recessive disorder, characterized by bleeding symptoms mainly associated with injury or surgery.

The initiation of blood coagulation involves tissue factor (TF)-induced allosteric activation of factor VIIa (FVIIa), which circulates in a zymogen-like state. In addition, the (most) active conformation of FVIIa presumably relies on a number of intramolecular interactions. We have characterized the role of Gly372(223) in FVIIa

The coagulation and fibrinolytic systems are central to the hemostatic mechanism, which works promptly on vascular injury and tissue damage. The rapid response is generated by specific molecular interactions between components in these systems. Thus, the regulation mechanism of the sys-tems is programmed in each component, as exemplified by the elegant pro-cesses in zymogen activation.
