Regulator of G-protein signaling (RGS) proteins were originally identified as negative regulators of G-protein-coupled receptor (GPCR) signaling via their GTPase-accelerating protein (GAP) activity. All RGS proteins contain evolutionarily conserved RGS domain; however, they differ in their size and regulatory domains. RGS1 and RGS10 are smaller than other RGS proteins, and their functions involve various inflammatory responses including autoimmune responses in both the periphery and the central nervous system (CNS). Neuroinflammation is the chronic inflammatory response in the CNS.

The transcription factor serum response factor (SRF) regulates expression of growth factor-dependent genes and muscle-specific genes in vertebrates. Homologous factors regulate differentiation of some ectodermic tissues in invertebrates. To explore the molecular basis of these different physiological functions, the functionality of human, Drosophila melanogaster and Artemia franciscana SRFs in mammalian cells has been compared in this article. D. melanogaster and, to a lesser extend, A.

The human immunodeficiency virus type 1, HIV-1, genome encodes a highly conserved regulatory gene product, Vpr (96 amino acids), which is incorporated into virions in quantities equivalent to those of the viral Gag protein. In infected cells, Vpr is believed to function during the early stages of HIV-1 replication (such as transcription of the proviral genome and migration of preintegration nuclear complex), blocks cells in G2 phase and triggers apoptosis.

The centromere–kinetochore complex is a highly specialized chromatin domain that both mediates and monitors chromosome–spindle interactions responsible for accurate partitioning of sister chromatids to daughter cells. Centromeres are distinguished from adjacent chromatin by specific patterns of histone modification and the presence of a centromere-specific histone H3 variant (e.g. CENP-A). Centromere-proximal regions usually correspond to sites of avid and persistent sister chromatid cohesion mediated by the conserved cohesin complex.

A technique has been developed to search a proteome database for new members of a functional class of membrane protein. It takes advantage of the highly conserved secondary structure of functionally related membrane proteins. Such proteins typically have the same number of transmembrane domains located at similar relative positions in their polypeptide sequence. This gives rise to a characteristic pattern of peaks in their hydropathy profiles.

Theb1,3-glycosyltransferase enzymes identi®ed to date share several conserved regions and conserved cysteine res-idues, all being located in the putative catalytic domain. To investigate the importance of these motifs and cysteines for the enzymatic activity, 14 mutants of the murine b1,3-galactosyltransferase-I gene were constructed and expressed in Sf9 insect cells. Seven mutations abolished the galacto-syltransferase activity.

Protein phosphatase 2A (PP2A) is an abundant heterotri-meric serine/threonine phosphatase containing highly con-served structural (A) and catalytic (C) subunits. Its diverse functions in the cell are determined by its association with a highly variable regulatory and targeting B subunit. At least three distinct gene families encoding B subunits are known: B/B55/CDC55, B¢/B56/RTS1 and B¢¢/PR72/130. No homologyhasbeen identi®edamong theBfamilies, and little is knownabout howtheseBsubunits interactwith the PP2A A and C subunits....

Previous structural andmutagenesis studies indicate that the invariant a-amino and a-carboxyl groups of glutamate receptor agonists are engaged in polar interactions with oppositely charged, conserved arginine and glutamate resi-dues in the ligand-binding domain ofa-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor. To examine the role of these residues (R507 and E727 in the GluR-D subunit) in the discrimination between agonists and anta-gonists, we analyzed the ligand-binding properties of homomeric GluR-D and its soluble ligand-binding domain withmutations at thesepositions....

We describe the structure and functional properties of an FTZ-F1 from the platyhelminth parasite of humans,Schistosoma mansoni, the first receptor from this family tobe characterized inaLophotrochozoan. It contains awell conservedDNA-binding domain (55–63%identity to other family members) and a poorly conserved ligand-binding domain (20% identity to that of zebrafish FF1a)....

The N-terminal part of Era contains a conserved GTPase domain, whereas the C-terminal part of the protein contains anRNA- andmembrane-binding domain, theKH domain. To investigate whether the binding of Era to 16S rRNA and membrane requires its GTPase activity and whether the GTPase domain is essential for these acti-vities, the N- and C-terminal parts of the Streptococ-cus pneumoniaeEra –Era-N(aminoacids 1–185) andEra-C (amino acids 141–299), respectively – were expressed and purified. ...

A conserved putative dimerization GxxxG motif located in the unique membrane-spanning segment of the ATP syn-thase subunit e was altered in yeast both by insertion of an alanine residue and by replacement of glycine by leucine residues. These alterations led to the loss of subunit gand the loss of dimeric and oligomeric forms of the yeast ATP syn-thase. Furthermore,as in null mutants devoid of either subunit e or subunit g,mitochondria displayed anomalous morphologies with onion-like structures. ...

Among the casp-ases is a subgroup whose primary function is to initiate apoptosis. Within their long prodomains, caspases-2, -9 and -12 contain a caspase activation and recruitment domain while caspases-8 and -10 bear death effector domains.

Oxysterol binding protein (OSBP) and its homologs have been shown to regulate lipid metabolism and vesicular transport. However, the exact molecular function of individual OSBP homologs remains uncharacterized. Here we demonstrate that the yeast OSBP homolog, Osh6p, bound phos-phatidic acid and phosphoinositides via its N-terminal half containing the conserved OSBP-related domain (ORD). Using a green fluorescent protein fusion chimera, Osh6p was found to localize to the cytosol and patch-like or punctate structures in the vicinity of the plasma membrane....

In humans, eight types of histone H1 exist (H1.1–H1.5, H1, H1t and H1oo), all consisting of a highly conserved globular domain and less con-served N- and C-terminal tails. Although the precise functions of these iso-forms are not yet understood, and H1 subtypes have been found to be dispensable for mammalian development, it is now clear that specific func-tions may be assigned to certain individual H1 subtypes.

Deviations in these canonical amino acids are observed in several putative adenylyl cyclase genes as apparent in several bacterial genomes. This suggests that a variety of catalytic mechanisms may actually exist.
