The central objective of the thesis "An economic analysis of the Australian Baby Bonus" is to examine the nature of the fertility response of Australian families to the financial incentive of the Baby Bonus. The efficacy of the policy in determining a change in both fertility and the timing of births is assessed by exploring the time series properties of national and Victorian fertility measures, while controlling for other possible determinants of fertility choice.

The current research is undertaken by performing Computational Fluid Dynamics (CFD) simulations for a 2D aerofoil fitted with a shock control bump at a transonic flow condition. Reynolds-Averaged Navier-Stokes equation coupled with turbulence models is adopted to solve the flowfield. The bump has a conventional geometric feature which consists of a ramp, a crest and a tail, and it has eight variants for which the designed parameters are reasonably specified according to the compressible aerodynamics theory.

The activity of outer membrane phospholipase A (OMPLA) is regulated by reversible dimerization. However, native OMPLA reconstituted in phospholipid vesicles was found to be present as a dimer but nevertheless inactive. To investigate the importance of dimerization for control of OMPLA activity, a covalent OMPLA dimer was constructed and its properties were compared to native OMPLA both in a micellar detergent and after reconstitution in a phospholipid bilayer.

Site-directedmutagenesis was used to investigate the control of 2-oxoacid cosubstrate selectivity by deacetoxycephalo-sporin C synthase. The wild-type enzyme has a requirement for 2-oxoglutarate and cannot efficiently use hydrophobic 2-oxoacids (e.g. 2-oxohexanoic acid, 2-oxo-4-methyl-penta-noic acid) as the cosubstrate. The followingmutant enzymes were produced: R258A, R258L, R258F, R258H and R258K. All of the mutants have broadened cosubstrate selectivity and were able to utilize hydrophobic 2-oxoacids.

Four pyruvate dehydrogenase kinase and two pyruvate dehydrogenase phosphatase isoforms function in adjusting the activation state of the pyruvate dehydrogenase complex (PDC) through determining the fraction of active (non-phosphorylated) pyruvate dehydrogenase component. The surface charge as well as the electrochemical properties and ligand binding abilities of the Gram-positive cell wall is controlled by the d-alanylation of the lipoteichoic acid. The incorporation of d-Ala into lipoteichoic acid requires the d-alanine:d-alanyl carrier protein ligase (DltA) and the carrier protein (DltC). We have heterologously expressed, purified, and assayed the substrate selectivity of the recombinant proteins DltA with its substrate DltC.

To facilitate the process of proteindesign and learn the basic rules that control the structure and stability of proteins, combinatorial methods have been developed to select or screen proteins with desired properties from libraries of mutants. One such method uses phage-display and proteo-lysis to select stably folded proteins. This method does not rely on specific properties of proteins for selection. There-fore, in principle it can be applied to any protein.

The control properties of biochemical pathways can be described by control coefficients and elasticities, as defined in the framework of metabolic control analysis. The determination of these parameters using the traditional metabolic control analysis relationships is, however, lim-ited by experimental difficulties (e.g. realizing and meas-uring small changes in biological systems) and lack of appropriate mathematical procedures (e.g. when the more practical large changes are made).

Model amphipathic peptides have been widely used as a tool to determine the structural and biological properties that control the interaction of pep-tides with membranes. Here, we have focused on the role of a central Pro in membrane-active peptides. To determine the role of Pro in structure, antibiotic activity, and interaction with phospholipids, we generated a ser-ies of model amphipathic a-helical peptides with different chain lengths and containing or lacking a single central Pro.
