For many years, the use of chemical agents to control crop pests has been degrading the environment, bringing problems to humans and all living things. An alternative to deal with the pests is the use of biopesticides, biological agents capable of controlling these harmful organisms.

Bacillus thuringiensis produces different insecticidal crystal proteins which are encoded by cry genes. Isolation was carried out from 17 diverse locations’ soil; nine bacterial isolates were separated and selected on the basis of presence of parasporal body. Isolate ABt 10 was screened for different cry genes with universal and gene specific primers.

Cry protein profiling of Bacillus thuringiensis isolated from different agro-climate soils of Punjab
Insect pests caused the yield loss in agricultural crops and forestry plants world widely. Commercial biopesticides based on Bacillus thuringiensis (Bt) have been used for pest management. The most important factor that makes Bt as commercial biopesticides is its remarkable ability to produce large quantities of larvicidal proteins (known as Cry protein or delta-endotoxin) that form crystalline inclusion bodies during sporulation.

Bacillus thuringiensis is produced crystalline proteins, these proteins were used in transgenic cotton to control insect pest. During present study, two trials were with 09 treatments plots of Bt cotton plants viz., 7, 14 and 21gm/m.sq. (T1, T2 amd T3) of nitrogen with one row combination of nBt cotton plants while T4, T5, T6 were having two rows combination of nBt cotton plants with same nitrogen treatment as in above mentioned plots whereas T7, T8 and T9 were only treatment with different concentration of nitrogen and there was no rows of nBt cotton plants around the plots were conducted.

Bacillus thuringiensis has a significant impact on biological pest control because of the insecticidal activity through its parasporal inclusion bodies (crystal proteins). Fig is an economically important plant in Turkey, agricultural pests result in a considerable economic loss in fig quality and cultivation.

a-Crystallin, a molecular chaperone and lens structural protein protects soluble enzymes against heat-induced aggregation and inactivation by a variety of molecules. In this study we investigated the chaperone function ofa-crys-tallininamorephysiological systeminwhicha-crystallinwas incorporated into red cell ghosts. Its ability to protect the intrinsic membrane protein Na/K-ATPase from external stresses was studied. Red cell ghosts were created by lysing the red cells and removing cytoplasmic contents by size-exclusion chromatography. ...

Several transcription factors with the function of setting the biological clock in vertebrates have been described. A detailed understanding of their nucleocytolasmic transport properties may uncover novel aspects of the regulation of the circadian rhythm. This assumption led us to perform a systematic analysis of the nuclear import characteristics of the different murine PER and CRY proteins, usingXenopusoocytes and HeLa cells as experimental systems.

The substrate specificity of the nuclear inclusion protein a (NIa) proteolytic enzymes from two potyviruses, the tobacco etch virus (TEV) and tobacco vein mottling virus (TVMV), was compared using oligopeptide substrates. Mutations were introduced into TEV protease in an effort to identify key determinants of substrate specificity. The specificity of the mutant enzymes was assessed by using peptides with complementary substitutions. The crys-tal structure of TEV protease and a homology model of TVMV protease were used to interpret the kinetic data....

A novel, type 1 ribosome-inactivating protein designated charybdin was isolated from bulbs of Charybdis maritimaagg. The protein, consisting of a single polypeptide chain with a molecular mass of 29 kDa, inhibited trans-lation in rabbit reticulocytes with an IC50of 27.2 nm. Plant genomic DNA extracted from the bulb was amplified by PCR between primers based on the N-terminal and C-terminal sequence of the protein from dissolved crys-tals.

The conformationally coupled mechanism by which ATP is utilized by yeast Hsp90 is now well characterized. In contrast, ATP utilization by human Hsp90s is less well studied, and appears to operate differently. To resolve these conflicting models, we have conducted a side-by-side biochem-ical analysis in a series of mutant yeast and human Hsp90s that have been both mechanistically and structurally characterized with regard to the crys-tal structure of the yeast Hsp90 protein.

The determination of the receptor-binding region of Cry toxins produced byBacillus thuringiensisis expected to facilitate an improvement in their insecticidal ability through protein engineering. We analyzed the region on Cry1Aa molecules involved in interactions with the cadherin-like protein receptor BtR175 using cysteine-substituted mutant toxins and several syn-thetic peptides corresponding to the loops in domain 2.

Numerous proteins cannot be sufficiently prepared by ordinary recombi-nant DNA techniques because they are unstable or have deleterious effects on the host cell. One idea to prepare such proteins is to produce them as protein inclusions. Here we developed a novel system to effectively prepare proteins by using peptide tags derived from the insecticidal Cry toxin of a soil bacterium, Bacillus thuringiensis.
