Assembly of the components of the thylakoidDpH-dependent/Tat protein transport machinery was analyzed in vitro. Upon incubation with intact chloroplasts, precur-sors to all three components, Hcf106, cpTatC and Tha4, were imported into the organelle and assembled into char-acteristic endogenous complexes. In particular, all of the imported cpTatC and approximately two-thirds of the imported Hcf106functionally assembled into 700 kDa complexes capable of binding Tat pathway precursor pro-teins.

Microfibrils and elastin are major constituents of elastic fibers, the assembly of which is dictated by multimolecular interactions.Microfibril-associated glycoprotein-1 (MAGP-1) is a microfibrillar component that interacts with the sol-uble elastin precursor, tropoelastin. We describe here the adaptation of a solid-phase binding assay that defines the effect of divalent cations on the interactions between MAGP-1 and tropoelastin. Using this assay, a strong cal-cium-dependent interaction was demonstrated, with a dis-sociation constant of 2.8 ± 0.3 nM, which fits a single-site binding model.

The occurrence of DNA double-strand breaks in the nucleus provokes in its structural organization a large-scale alteration whose molecular basis is still mostly unclear. Here, we show that double-strand breaks trigger pref-erential assembly of nucleoproteins in human cellular fractions and that they mediate the separation of large protein–DNA aggregates from aque-ous solution.
