The new antigen receptor (IgNAR) from sharks is a disul-phide bonded dimer of two protein chains,each containing one variable and five constant domains,and functions as an antibody. This study describes the construction of a library of single-chain antibody fragments (scFvs) from a single human donor by individual amplification of all heavy and light variable domains (1.1·10 8 recombinants). The lib-rary was panned using the phage display technique, which allowed selection of specific scFvs (3F and C1) capable of recognizing Cn2, the major toxic component of Centruroides noxiusscorpion venom.

This motif is widely distributed in eukaryotic proteins, as well as in proteins from bac-teria and viruses. dsRBM-containing proteins are involved in processes ran-ging from RNA editing to protein phosphorylation in translational control and contain a variable number of dsRBM domains.

Intracellularly expressed antibodies (intrabodies) have been used to inhibit the function of various kinds of protein inside cells. However, problems with stability and functional expression of intrabodies in the cytosol remain unsolved. Recurrent missense fibroblast growth factor receptor 3 (FGFR3) mutations have been ascribed to skeletal dysplasias of variable severity including the lethal neonatal thanatophoric dysplasia types I (TDI) and II (TDII). Vertebrate metallothioneins are found to contain Zn(II) and variable amounts of Cu(I),in vivo, and are believed to be important for d 10 -metal control. To date, structural information is available for the Zn(II) and Cd(II) forms, but not for the Cu(I) or mixed metal forms. Cu(I) binding to metallothionein-1 has been investigated by circular dichroism, luminescence and 1 H NMR using two synthetic fragments representing thea- and the b-domain.
